FISH has gained an irreplaceable place in microbiology because of its ability to detect and locate a microorganism, or a group of organisms, within complex samples. However, FISH role has evolved drastically in the last few decades and its value has been boosted by several advances in signal intensity, imaging acquisitions, automation, method robustness, and, thus, versatility. This has resulted in a range of FISH variants that gave researchers the ability to access a variety of other valuable information such as complex population composition, metabolic activity, gene detection/quantification, or subcellular location of genetic elements. In this chapter, we will review the more relevant FISH variants, their intended use, and how they address particular challenges of classical FISH. Itowolo is a fishing village being boarded with Lagos lagoon on nearly every side except the southern part which opened to the Ikorodu road from Mile 12 part of Agboyi - Ketu Local Council Development Area. During the raining season, the water level increases which possibly cause flood at heavy down pour while low water level of higher salinity during the dry season. The indigenes are primarily fisherfolks and they are effective in their fishing activities by having individual functional canoe ranging from small unmotorised paddled dugout type to outboard ones. The main gears used for fishing in the area are cast nets, gill nets, traps, hooks and lines among which cast net is prominently used Their landings during the raining season involve different species of fishes such as Heterobranchus species, Heterotis species, Channa species including others while tilapia of different species take the greatest percentage of their catches during the dry season. The shrimps top the population of catches between the months of July September. The dwellers during off fishing season engage in other economic activities that generate income such as petty trading, palm tapping, fuel wood trading, transport business, mat making, hairdressing, fashion designing, computer business centres and telecommunication vendors. In general, fishing activities are characterized by low unit productivity, intensive labour, relative low capital, low operational costs and limited gear coverage. The Florida Fish and Wildlife Conservation Commission (FWC) collected annual trawl data at 27 open-water sites from 1987 to 1991 (Bull et al. 1995). Nearly 37,000 fish were recorded in 438 10-minute open-water trawls (Bull et al. 1995). Seven species accounted for 98% of the total number and total fish biomass. Clustering of sites based on mean catch of the primary species expressed as number and weight produced four distinct groups. The groups were labeled as the northeast shore, northwest shore, south-southwest shore and open water area. Areal fish distribution patterns also were compared using analysis of variance (ANOVA) and Tukeyâ s HSD post hoc test. Within the four groups there were significant differences in the distribution of certain fish species. In addition to the open-water trawl sites, the FWC has utilized electrofishing techniques to collect annual largemouth bass (Micropterous salmodies) (LMB) data from 22 near-shore and interior marsh locations since 1999 (Havens et al. 2004). Although the trawl and electrofishing data provide some baseline information, still there is limited data regarding temporal changes in the community structure, density and condition of the primary sport fish LMB, black crappie (Pomoxis nigromaculatus), bluegill (Lepomis macrochirus) and redear (Lepomis microlophus) sunfish) and other fish species in Lake Okeechobee. During this study, fish species will be collected from 49 historic sampling locations. Fish assemblages in the 27 open water regions of the lake will be sampled with an Otter Trawl net. The 22 near-shore and interior marsh sites will be sampled utilizing electrofishing gear. Ancillary data, including water temperature, dissolved oxygen, pH, turbidity, conductivity, and sediment/aquatic plant type will be recorded at the 49 sampling locations. The two historic sets of non-MAP data will be used to help establish baseline conditions for the near-shore, interior marsh and open-water fishery. It is appropriate to include the non-MAP data in our analysis as current sampling will occur at the historical locations and sampling methods will be similar. We anticipate significant spatial differences in fish abundance and biomass will exist at the near-shore, interior marsh and open water sites. Therefore, similar statistical tests including cluster analysis and analysis of variance should be used to evaluate temporal changes in the near-shore and open water fishery. Detailed statistical analysis should be conducted at a minimum of every three years to evaluate long-term trends and establish relationships between fish distribution, condition, and community structure and environmental conditions including habitat and water depth. The objectives of this project are to evaluate temporal changes in Lake Okeechobeeâ s fishery by determining annual changes in the areal distribution, condition, density and community structure (year classes) of all major fish species found in the near-shore, interior marsh and open-water regions of the lake. Ancillary data including water temperature, dissolved oxygen, pH, turbidity, conductivity, and sediment type also will be recorded.

The cod was tagged in the Southern North Sea on 25/03/2005 and was recaptured in the Eastern English Channel on the 09/01/2006. The cod migrates down through the English Channel, resides in the Celtic Sea and then returns. The dataset contains 291 days of information. Each day consists of 144 vertical movement rates (in metres), one horizontal movement rate (in kilometres) and one turning angle (in radians, from -pi to pi). Vertical movement rates have been log (natural log) transformed. A description of tag type, fish capture, tag attachment and tag accuracy can be found in Righton et al. (2010). Righton et al. (2010) is published in Marine Ecology Progress Series and is freely available here http://www.int-res.com/articles/feature/m420p001.pdf. The data storage tag was pre-programmed to record depth (m) at 10-minute intervals for the duration of deployment. Vertical movement rates were calculated as the absolute difference between corresponding 10-minute depth measurements summed to the daily level. Horizontal movement rates were calculated in a two-step approach. First, daily geolocations were estimated via a Fokker-Planck-based method that combines a tidal location method and a Bayesian state-space model (see Pedersen et al. 2008 for model details). Second, the straight-line distance between daily geolocations is calculated using the Great Circle equation. Turning angles between successive geolocations were also calculated. Pedersen et al. Itowolo is a fishing village being boarded with Lagos lagoon on nearly every side except the southern part which opened to the Ikorodu road from Mile 12 part of Agboyi - Ketu Local Council Development Area. During the raining season, the water level increases which possibly cause flood at heavy down pour while low water level of higher salinity during the dry season. The indigenes are primarily fisherfolks and they are effective in their fishing activities by having individual functional canoe ranging from small unmotorised paddled dugout type to outboard ones. The main gears used for fishing in the area are cast nets, gill nets, traps, hooks and lines among which cast net is prominently used Their landings during the raining season involve different species of fishes such as Heterobranchus species, Heterotis species, Channa species including others while tilapia of different species take the greatest percentage of their catches during the dry season. The shrimps top the population of catches between the months of July September. The dwellers during off fishing season engage in other economic activities that generate income such as petty trading, palm tapping, fuel wood trading, transport business, mat making, hairdressing, fashion designing, computer business centres and telecommunication vendors. In general, fishing activities are characterized by low unit productivity, intensive labour, relative low capital, low operational costs and limited gear coverage. The Florida Fish and Wildlife Conservation Commission (FWC) collected annual trawl data at 27 open-water sites from 1987 to 1991 (Bull et al. 1995). Nearly 37,000 fish were recorded in 438 10-minute open-water trawls (Bull et al. 1995). Seven species accounted for 98% of the total number and total fish biomass. Clustering of sites based on mean catch of the primary species expressed as number and weight produced four distinct groups. The groups were labeled as the northeast shore, northwest shore, south-southwest shore and open water area. Areal fish distribution patterns also were compared using analysis of variance (ANOVA) and Tukeyâ s HSD post hoc test. Within the four groups there were significant differences in the distribution of certain fish species. In addition to the open-water trawl sites, the FWC has utilized electrofishing techniques to collect annual largemouth bass (Micropterous salmodies) (LMB) data from 22 near-shore and interior marsh locations since 1999 (Havens et al. 2004). Although the trawl and electrofishing data provide some baseline information, still there is limited data regarding temporal changes in the community structure, density and condition of the primary sport fish LMB, black crappie (Pomoxis nigromaculatus), bluegill (Lepomis macrochirus) and redear (Lepomis microlophus) sunfish) and other fish species in Lake Okeechobee. During this study, fish species will be collected from 49 historic sampling locations. Fish assemblages in the 27 open water regions of the lake will be sampled with an Otter Trawl net. The 22 near-shore and interior marsh sites will be sampled utilizing electrofishing gear. Ancillary data, including water temperature, dissolved oxygen, pH, turbidity, conductivity, and sediment/aquatic plant type will be recorded at the 49 sampling locations. The two historic sets of non-MAP data will be used to help establish baseline conditions for the near-shore, interior marsh and open-water fishery. It is appropriate to include the non-MAP data in our analysis as current sampling will occur at the historical locations and sampling methods will be similar. We anticipate significant spatial differences in fish abundance and biomass will exist at the near-shore, interior marsh and open water sites. Therefore, similar statistical tests including cluster analysis and analysis of variance should be used to evaluate temporal changes in the near-shore and open water fishery. Detailed statistical analysis should be conducted at a minimum of every three years to evaluate long-term trends and establish relationships between fish distribution, condition, and community structure and environmental conditions including habitat and water depth. The objectives of this project are to evaluate temporal changes in Lake Okeechobeeâ s fishery by determining annual changes in the areal distribution, condition, density and community structure (year classes) of all major fish species found in the near-shore, interior marsh and open-water regions of the lake. Ancillary data including water temperature, dissolved oxygen, pH, turbidity, conductivity, and sediment type also will be recorded.
