    POU genes are a family of evolutionarily conserved transcription factors with key functions in cell type specification and neurogenesis. In vitro experiments have indicated that the expression of some POU genes is controlled by the intercellular signaling molecule retinoic acid (RA). In this work, we aimed to characterize the roles of RA signaling in the regulation of POU genes in vivo. To do so, we studied POU genes during the development of the cephalochordate amphioxus, an animal model crucial for understanding the evolutionary origins of vertebrates. The expression patterns of amphioxus POU genes were assessed at different developmental stages by chromogenic in situ hybridization and hybridization chain reaction. Expression was further assessed in embryos subjected to pharmacological manipulation of endogenous RA signaling activity. In addition to a detailed description of the effects of these treatments on amphioxus POU gene expression, our survey included the first description of Pou2 and Pou6 expression in amphioxus embryos. We found that Pit-1, Pou2, Pou3l, and Pou6 expression are not affected by alterations of endogenous RA signaling levels. In contrast, our experiments indicated that Brn1/2/4 and Pou4 expression are regulated by RA signaling in the endoderm and the nerve cord, respectively. The effects of the treatments on Pou4 expression in the nerve cord revealed that, in developing amphioxus, RA signaling plays a dual role by (1) providing anteroposterior patterning information to neural cells and (2) specifying neural cell types. This finding is coherent with a terminal selector function of Pou4 for GABAergic neurons in amphioxus and represents the first description of RA-induced changes in POU gene expression in vivo.

    The AlpArray programme is a multinational, European consortium to advance our understanding of orogenesis and its relationship to mantle dynamics, plate reorganizations, surface processes and seismic hazard in the Alps–Apennines–Carpathians–Dinarides orogenic system. The AlpArray Seismic Network has been deployed with contributions from 36 institutions from 11 countries to map physical properties of the lithosphere and asthenosphere in 3D and thus to obtain new, high-resolution geophysical images of structures from the surface down to the base of the mantle transition zone. With over 600 broadband stations operated for 2 years, this seismic experiment is one of the largest simultaneously operated seismological networks in the academic domain, employing hexagonal coverage with station spacing at less than 52 km. This dense and regularly spaced experiment is made possible by the coordinated coeval deployment of temporary stations from numerous national pools, including ocean-bottom seismometers, which were funded by different national agencies. They combine with permanent networks, which also required the cooperation of many different operators. Together these stations ultimately fill coverage gaps. Following a short overview of previous large-scale seismological experiments in the Alpine region, we here present the goals, construction, deployment, characteristics and data management of the AlpArray Seismic Network, which will provide data that is expected to be unprecedented in quality to image the complex Alpine mountains at depth.     Mayfly (Ephemeroptera) systematics has considerably changed over the years, but many questions have yet to be answered. The synergistic connection between traditional knowledge and new data sources, producing increasingly complex information, has become a compelling issue for modern taxonomy. Molecular tests and the use of reliable reference sequence libraries may constitute effective complements to the traditional method in guiding recognition of species and giving information about taxonomic incongruences which require further examination. In the present study, we sought to verify the current Italian mayfly nomenclatural system through DNA barcoding and relevant points to reliably manage the available amount of morpho-ecological and molecular data are discussed. We investigated COI (Cytochrome oxidase I) sequence variation in 163 individuals of Italian mayflies, 126 of which were previously assigned to 24 morphologically recognised species, and 37 could be attributed only to generic taxonomic entities (“sp.”, “cf.” or “gr.”). DNA barcoding statistical tests for species delimitation hypotheses based on genetic distances and inferred gene trees were integrated with GenBank searches and surveys of the historical literature to better understand the knowledge acquired on the status and diversity of the investigated taxa. Combined criteria to define three categories of reliability were then assessed. Concurrent data allowing unambiguous identification were attained for only eight species. High intraspecific genetic distances (> 3%) and a lack of reliable reference material or convincing taxonomic information evidenced 29 critical states, deserving further investigation. Solid species names, potential cryptic species and entities about which little is known are pointed out for a future upgrade/reorganisation of the taxonomy of Italian Ephemeroptera.

    During the austral summer of 2014, an oceanographic cruise was conducted in the Ross Sea in the framework of the RoME (Ross Sea Mesoscale Experiment) Project. Forty-three hydrological stations were sampled within three different areas: the northern Ross Sea (RoME 1), Terra Nova Bay (RoME 2), and the southern Ross Sea (RoME 3). The ecological and photophysiological characteristics of the phytoplankton were investigated (i.e., size structure, functional groups, PSII maximum quantum efficiency, photoprotective pigments), as related to hydrographic and chemical features. The aim was to identify the mechanisms that modulate phytoplankton blooms, and consequently, the fate of organic materials produced by the blooms. The observed biomass standing stocks were very high (e.g., integrated chlorophyll-a up to 371 mg m-2 in the top 100 m). Large differences in phytoplankton community composition, relative contribution of functional groups and photosynthetic parameters were observed among the three subsystems. The diatoms (in different physiological status) were the dominant taxa in RoME 1 and RoME 3; in RoME 1, a post-bloom phase was identified, whereas in RoME 3, an active phytoplankton bloom occurred. In RoME 2, diatoms co-occurred with Phaeocystis antarctica, but were vertically segregated by the upper mixed layer, with senescent diatoms dominating in the upper layer, and P. antarctica blooming in the deeper layer. The dominance of the phytoplankton micro-fraction over the whole area and the high Chl-a suggested the prevalence of non-grazed large cells, independent of the distribution of the two functional groups. These data emphasise the occurrence of significant temporal changes in the phytoplankton biomass in the Ross Sea during austral summer. The mechanisms that drive such changes and the fate of the carbon production are probably related to the variations in the limiting factors induced by the concurrent hydrological modifications to the Ross Sea, and they remain to be fully clarified. The comparison of conditions observed during summer 2014 and those reported for previous years reveal considerably different ecological assets that might be the result of current climate change. This suggests that further changes can be expected in the future, even at larger oceanic scales.

    Abstract This study focuses on the potential explanations for a Phaeocystis antarctica summer bloom occurred in stratified waters of Terra Nova Bay (TNB) - which is part of the Antarctic Special Protected Area (n.161) in the Ross Sea - trough a multi-parameter correlative approach. Many previous studies have highlighted that water column stratification typically favors diatom dominance compared to the colonial haptophyte P. antarctica, in the Ross Sea, and this correlation has often been used to explain the historic dominance of diatoms in TNB. To explore the spatial and temporal progression of P. antarctica bloom in coastal waters, four stations were sampled three times each between December 31, 2009 and January 13, 2010. Taxonomic and pigment composition of phytoplankton communities, macro-nutrient concentrations and various different indices, all indicated the relative dominance of P. antarctica. Cell abundances revealed that P. antarctica contributed 79% of total cell counts in the upper 25 m and 93% in the lower photic zone. Similarly, a strong correlation was observed between Chl-a and the Hex:Fuco pigment ratio, corroborating the microscopic analyses. Recent studies have shown that iron can trigger colonial P. antarctica blooms. Based on the Hex:Chl-c3 proxy for iron limitation in P. antarctica, we hypothesize that anomalously higher iron fluxes were responsible for the unusual bloom of colonial P. antarctica observed in TNB.

    Despite the discovery and development of novel therapies, cancer is still a leading cause of death worldwide. In order to grow, tumor cells require large quantities of nutrients involved in metabolic processes, and an increase in iron levels is known to contribute to cancer proliferation. Iron plays an important role in the active site of a number of proteins involved in energy metabolism, DNA synthesis and repair, such as ribonucleotide reductase, which induce G0/S phase arrest and exert a marked antineoplastic effect, particularly in leukemia and neuroblastoma. Iron-depletion strategies using iron chelators have been shown to result in cell cycle arrest and apoptosis. Deferoxamine (DFO) was the first FDA-approved drug for the treatment of iron overload pathologies, and has also been recognized as having anticancer properties. The high cost, low permeability and short plasma half-life of DFO led to the development of other iron-chelating drugs. Pyridoxal isonicotinoyl hydrazone (PIH) and its analogs chelate cellular iron by tridentate binding, and inhibit DNA synthesis more robustly than DFO, demonstrating an effective antiproliferative activity. Here, we investigated the biological effects of a PIH derivative, 3-chloro-N′-(2-hydroxybenzylidene)benzohydrazide (CHBH), known to be a lysine-specific histone demethylase 1A inhibitor. We showed that CHBH is able to induce cell proliferation arrest in several human cancer cell lines, including lung, colon, pancreas and breast cancer, at micromolar levels. Our findings indicate that CHBH exerts a dual anticancer action by strongly impairing iron metabolism and modulating chromatin structure and function.     The new benthic toxic dinoflagellate, Ostreopsis fattorussoi sp. nov., is described from the Eastern Mediterranean Sea, Lebanon and Cyprus coasts, and is supported by morphological and molecular data. The plate formula, Po, 3′, 7″, 6c, 7s, 5‴, 2′′′′, is typical for the Ostreopsis genus. It differs from all other Ostreopsis species in that (i) the curved suture between plates 1′ and 3′ makes them approximately hexagonal, (ii) the 1′ plate lies in the left half of the epitheca and is obliquely orientated leading to a characteristic shape of plate 6″. The round thecal pores are bigger than the other two Mediterranean species (O. cf. ovata and O. cf. siamensis). O. fattorussoi is among the smallest species of the genus (DV: 60.07 ± 5.63 μm, AP: 25.66 ± 2.97 μm, W: 39.81 ± 5.05 μm) along with O. ovata. Phylogenetic analyses based on the LSU and internal transcribed spacer rDNA shows that O. fattorussoi belongs to the Atlantic/Mediterranean Ostreopsis spp. clade separated from the other Ostreopsis species. Ostreopsis fattorussoi produces OVTX‐a and structural isomers OVTX‐d and ‐e, O. cf. ovata is the only other species of this genus known to produce these toxins. The Lebanese O. fattorussoi did not produce the new palytoxin‐like compounds (ovatoxin‐i, ovatoxin‐j1, ovatoxin‐j2, and ovatoxin‐k) that were previously found in O. fattorussoi from Cyprus. The toxin content was in the range of 0.28–0.94 pg · cell−1. On the Lebanon coast, O. fattorussoi was recorded throughout the year 2015 (temperature range 18°C–31.5°C), with peaks in June and August.

    POU genes are a family of evolutionarily conserved transcription factors with key functions in cell type specification and neurogenesis. In vitro experiments have indicated that the expression of some POU genes is controlled by the intercellular signaling molecule retinoic acid (RA). In this work, we aimed to characterize the roles of RA signaling in the regulation of POU genes in vivo. To do so, we studied POU genes during the development of the cephalochordate amphioxus, an animal model crucial for understanding the evolutionary origins of vertebrates. The expression patterns of amphioxus POU genes were assessed at different developmental stages by chromogenic in situ hybridization and hybridization chain reaction. Expression was further assessed in embryos subjected to pharmacological manipulation of endogenous RA signaling activity. In addition to a detailed description of the effects of these treatments on amphioxus POU gene expression, our survey included the first description of Pou2 and Pou6 expression in amphioxus embryos. We found that Pit-1, Pou2, Pou3l, and Pou6 expression are not affected by alterations of endogenous RA signaling levels. In contrast, our experiments indicated that Brn1/2/4 and Pou4 expression are regulated by RA signaling in the endoderm and the nerve cord, respectively. The effects of the treatments on Pou4 expression in the nerve cord revealed that, in developing amphioxus, RA signaling plays a dual role by (1) providing anteroposterior patterning information to neural cells and (2) specifying neural cell types. This finding is coherent with a terminal selector function of Pou4 for GABAergic neurons in amphioxus and represents the first description of RA-induced changes in POU gene expression in vivo.

    The AlpArray programme is a multinational, European consortium to advance our understanding of orogenesis and its relationship to mantle dynamics, plate reorganizations, surface processes and seismic hazard in the Alps–Apennines–Carpathians–Dinarides orogenic system. The AlpArray Seismic Network has been deployed with contributions from 36 institutions from 11 countries to map physical properties of the lithosphere and asthenosphere in 3D and thus to obtain new, high-resolution geophysical images of structures from the surface down to the base of the mantle transition zone. With over 600 broadband stations operated for 2 years, this seismic experiment is one of the largest simultaneously operated seismological networks in the academic domain, employing hexagonal coverage with station spacing at less than 52 km. This dense and regularly spaced experiment is made possible by the coordinated coeval deployment of temporary stations from numerous national pools, including ocean-bottom seismometers, which were funded by different national agencies. They combine with permanent networks, which also required the cooperation of many different operators. Together these stations ultimately fill coverage gaps. Following a short overview of previous large-scale seismological experiments in the Alpine region, we here present the goals, construction, deployment, characteristics and data management of the AlpArray Seismic Network, which will provide data that is expected to be unprecedented in quality to image the complex Alpine mountains at depth.     Mayfly (Ephemeroptera) systematics has considerably changed over the years, but many questions have yet to be answered. The synergistic connection between traditional knowledge and new data sources, producing increasingly complex information, has become a compelling issue for modern taxonomy. Molecular tests and the use of reliable reference sequence libraries may constitute effective complements to the traditional method in guiding recognition of species and giving information about taxonomic incongruences which require further examination. In the present study, we sought to verify the current Italian mayfly nomenclatural system through DNA barcoding and relevant points to reliably manage the available amount of morpho-ecological and molecular data are discussed. We investigated COI (Cytochrome oxidase I) sequence variation in 163 individuals of Italian mayflies, 126 of which were previously assigned to 24 morphologically recognised species, and 37 could be attributed only to generic taxonomic entities (“sp.”, “cf.” or “gr.”). DNA barcoding statistical tests for species delimitation hypotheses based on genetic distances and inferred gene trees were integrated with GenBank searches and surveys of the historical literature to better understand the knowledge acquired on the status and diversity of the investigated taxa. Combined criteria to define three categories of reliability were then assessed. Concurrent data allowing unambiguous identification were attained for only eight species. High intraspecific genetic distances (> 3%) and a lack of reliable reference material or convincing taxonomic information evidenced 29 critical states, deserving further investigation. Solid species names, potential cryptic species and entities about which little is known are pointed out for a future upgrade/reorganisation of the taxonomy of Italian Ephemeroptera.

    During the austral summer of 2014, an oceanographic cruise was conducted in the Ross Sea in the framework of the RoME (Ross Sea Mesoscale Experiment) Project. Forty-three hydrological stations were sampled within three different areas: the northern Ross Sea (RoME 1), Terra Nova Bay (RoME 2), and the southern Ross Sea (RoME 3). The ecological and photophysiological characteristics of the phytoplankton were investigated (i.e., size structure, functional groups, PSII maximum quantum efficiency, photoprotective pigments), as related to hydrographic and chemical features. The aim was to identify the mechanisms that modulate phytoplankton blooms, and consequently, the fate of organic materials produced by the blooms. The observed biomass standing stocks were very high (e.g., integrated chlorophyll-a up to 371 mg m-2 in the top 100 m). Large differences in phytoplankton community composition, relative contribution of functional groups and photosynthetic parameters were observed among the three subsystems. The diatoms (in different physiological status) were the dominant taxa in RoME 1 and RoME 3; in RoME 1, a post-bloom phase was identified, whereas in RoME 3, an active phytoplankton bloom occurred. In RoME 2, diatoms co-occurred with Phaeocystis antarctica, but were vertically segregated by the upper mixed layer, with senescent diatoms dominating in the upper layer, and P. antarctica blooming in the deeper layer. The dominance of the phytoplankton micro-fraction over the whole area and the high Chl-a suggested the prevalence of non-grazed large cells, independent of the distribution of the two functional groups. These data emphasise the occurrence of significant temporal changes in the phytoplankton biomass in the Ross Sea during austral summer. The mechanisms that drive such changes and the fate of the carbon production are probably related to the variations in the limiting factors induced by the concurrent hydrological modifications to the Ross Sea, and they remain to be fully clarified. The comparison of conditions observed during summer 2014 and those reported for previous years reveal considerably different ecological assets that might be the result of current climate change. This suggests that further changes can be expected in the future, even at larger oceanic scales.

    Abstract This study focuses on the potential explanations for a Phaeocystis antarctica summer bloom occurred in stratified waters of Terra Nova Bay (TNB) - which is part of the Antarctic Special Protected Area (n.161) in the Ross Sea - trough a multi-parameter correlative approach. Many previous studies have highlighted that water column stratification typically favors diatom dominance compared to the colonial haptophyte P. antarctica, in the Ross Sea, and this correlation has often been used to explain the historic dominance of diatoms in TNB. To explore the spatial and temporal progression of P. antarctica bloom in coastal waters, four stations were sampled three times each between December 31, 2009 and January 13, 2010. Taxonomic and pigment composition of phytoplankton communities, macro-nutrient concentrations and various different indices, all indicated the relative dominance of P. antarctica. Cell abundances revealed that P. antarctica contributed 79% of total cell counts in the upper 25 m and 93% in the lower photic zone. Similarly, a strong correlation was observed between Chl-a and the Hex:Fuco pigment ratio, corroborating the microscopic analyses. Recent studies have shown that iron can trigger colonial P. antarctica blooms. Based on the Hex:Chl-c3 proxy for iron limitation in P. antarctica, we hypothesize that anomalously higher iron fluxes were responsible for the unusual bloom of colonial P. antarctica observed in TNB.

    Despite the discovery and development of novel therapies, cancer is still a leading cause of death worldwide. In order to grow, tumor cells require large quantities of nutrients involved in metabolic processes, and an increase in iron levels is known to contribute to cancer proliferation. Iron plays an important role in the active site of a number of proteins involved in energy metabolism, DNA synthesis and repair, such as ribonucleotide reductase, which induce G0/S phase arrest and exert a marked antineoplastic effect, particularly in leukemia and neuroblastoma. Iron-depletion strategies using iron chelators have been shown to result in cell cycle arrest and apoptosis. Deferoxamine (DFO) was the first FDA-approved drug for the treatment of iron overload pathologies, and has also been recognized as having anticancer properties. The high cost, low permeability and short plasma half-life of DFO led to the development of other iron-chelating drugs. Pyridoxal isonicotinoyl hydrazone (PIH) and its analogs chelate cellular iron by tridentate binding, and inhibit DNA synthesis more robustly than DFO, demonstrating an effective antiproliferative activity. Here, we investigated the biological effects of a PIH derivative, 3-chloro-N′-(2-hydroxybenzylidene)benzohydrazide (CHBH), known to be a lysine-specific histone demethylase 1A inhibitor. We showed that CHBH is able to induce cell proliferation arrest in several human cancer cell lines, including lung, colon, pancreas and breast cancer, at micromolar levels. Our findings indicate that CHBH exerts a dual anticancer action by strongly impairing iron metabolism and modulating chromatin structure and function.     The new benthic toxic dinoflagellate, Ostreopsis fattorussoi sp. nov., is described from the Eastern Mediterranean Sea, Lebanon and Cyprus coasts, and is supported by morphological and molecular data. The plate formula, Po, 3′, 7″, 6c, 7s, 5‴, 2′′′′, is typical for the Ostreopsis genus. It differs from all other Ostreopsis species in that (i) the curved suture between plates 1′ and 3′ makes them approximately hexagonal, (ii) the 1′ plate lies in the left half of the epitheca and is obliquely orientated leading to a characteristic shape of plate 6″. The round thecal pores are bigger than the other two Mediterranean species (O. cf. ovata and O. cf. siamensis). O. fattorussoi is among the smallest species of the genus (DV: 60.07 ± 5.63 μm, AP: 25.66 ± 2.97 μm, W: 39.81 ± 5.05 μm) along with O. ovata. Phylogenetic analyses based on the LSU and internal transcribed spacer rDNA shows that O. fattorussoi belongs to the Atlantic/Mediterranean Ostreopsis spp. clade separated from the other Ostreopsis species. Ostreopsis fattorussoi produces OVTX‐a and structural isomers OVTX‐d and ‐e, O. cf. ovata is the only other species of this genus known to produce these toxins. The Lebanese O. fattorussoi did not produce the new palytoxin‐like compounds (ovatoxin‐i, ovatoxin‐j1, ovatoxin‐j2, and ovatoxin‐k) that were previously found in O. fattorussoi from Cyprus. The toxin content was in the range of 0.28–0.94 pg · cell−1. On the Lebanon coast, O. fattorussoi was recorded throughout the year 2015 (temperature range 18°C–31.5°C), with peaks in June and August.
