    Water Quality issues in many Pacific countries are rising, with the increase in coastal populations and associated urban runoff but management requires contamination issues in the aquatic environment to be identified and prioritised. In Vanuatu and Solomon Islands there are few laboratories and resources to assess for the presence or impact of complex chemical contaminants. The extent and impact of chemical contamination of the marine and coastal environment is poorly described. Passive chemical samplers were used to measure a range of aquatic pollutants around the capital cities, Honiara (Solomon Islands) and Port Vila (Vanuatu). We detected a range of chemicals indicative of agricultural and industrial contamination and a few sites had concerning concentrations of specific hydrocarbons and pesticides. The rapid ecotoxicology test, Microtox, indicated toxic impacts in rivers, coastal sites and urban drains This work provides new data on chemical contamination and possible impacts of that contamination for both countries. The techniques could be applied widely across the region to generate critical data for environmental management, guide monitoring efforts and measure the impact of policy or land-use changes.

    Abstract In the Southwest Pacific Ocean, waters transit from the subtropical gyre before being redistributed equatorward and poleward. While the mean pathways are known, the contribution to the mixing and transport of the water from mesoscale eddies has not been comprehensively investigated. In this research, satellite altimetry data, combined with an eddy detection and tracking algorithm is used to investigate the distribution and surface characteristics of mesoscale eddies in this region of complex bathymetry (10°S–30°S, 140°E–190°E). Detected eddies are then colocalized with in situ data from Argo floats to determine their vertical structure and the effect of eddies on the water masses. The numerous islands affect the eddy behavior as most eddies are formed in the lee of islands, propagate westward and decay when encountering shallow bathymetry. Eddies are sparse and short‐lived in the tropical area north of Fiji, impacting only the top 200 meters of water. They do not appear to be able to trap and transport waters in this region. In the Coral Sea, a region of lateral shear between currents transporting waters of different origins, eddies are more numerous and energetic. They affect the water properties down to at least 500 m depth, and anticyclonic eddies trap water to ∼200 m, contributing to the upper thermocline waters mixing and transport. South of New Caledonia, mesoscale eddies are ubiquitous, with typical lifetimes longer than 5 months. They affect the temperature, salinity, and velocities down to ∼1,000 m depth, and weakly contribute to the mixing of lower thermocline waters. International audience

    Heavy rain events alter the biogeochemical outflows, affects water quality and ecosystem health within the coastal waters of small Pacific Islands. We characterized snapshots of the optical fingerprints of dissolved organic matter (DOM) sources together with the select nutrients, biogeochemical and physical variables for 10 stations in December 2017 in Laucala Bay, Fiji Islands. DOM absorption coefficients and fluorescence components were determined via spectrofluorometry and Parallel Factor Analysis identifying four components: Type M, two terrestrial (humic, fulvic) components, and a protein component linked to marine biological activity. Associations of DOM together with climate variables and the other tested variables were determined via principal component, hierarchical cluster, and cross-correlation (Pearson) analysis. All component s (together with most tested variables) displayed higher values (plumes) at the southwest coast consistent with surface currents outflow during the wet season. Type M component associated with two allochthonous fluorescent components signaling anthropogenic forcings via riverine outflows. Terrigenous inputs association with autochthonous chromophoric dissolved organic matter (CDOM) is indicative of tidal mixing, dilution, and bottom resuspension processes. Positive correlations of dissolved organic carbon (DOC) with nutrients (NOx, PO4) elucidates DOM being utilized as energy sources. The positive correlation of DON with nutrients (NOx, PO4, Si(OH)4) reflects the role of DON as a nutrient source consistent with chlorophyll plume formation International audience

    There has been a growing interest in medicinal plants by the locals of the Pacific Islands since it is readily available. Annona muricata, commonly called soursop is one of such plants which is popular because it is being used to treat initial stages of cancer due to its antioxidant properties. This study investigated various extraction methods which could be used to compare and see the effectiveness of each on the chromosomes. The cytotoxicity and genotoxicity effect were studied using Allium cepa root tip cells. Eight onion bulbs were subjected to the treatment groups which were made using methanol, water and hexane extracts of A. muricata. Three different concentrations from each extraction solvent were used. The A. cepa root tip cells were subjected for 24 h to three different leaf extractions after left to grow in water. Concentrations of 12.5, 25 and 50 g/L were used. After the treatment, onion bulbs were put back in water to recover. Hence, 5 root tips from each bulb were removed (before treatment, treatment and recovery) with cells undergoing mitosis counted under 400x of a compound microscope. All studied concentrations of A. muricata methanol extract showed significant (p≤0.05) drop in the mean mitotic index when compared with control (mean MI reduced by 9.9, 44.4 and 54.2% for 12.5, 25 and 50 g/L respectively). A. muricata water extract also showed significant differences at 12.5 g/L where mitotic index (MI) was reduced by 8.6%. At 25 g/L, the reduction was by 35.3% and by 42.8% at 50 g/L. Drastic effect (p≤0.05) was observed with the soursop-hexane extract that reduced the MI significantly in 12.5 g/L by 5.0% and by 29.7% at 25 g/L. A huge reduction in MI was observed at 50 g/L where MI declined by 72.8%. Chromosomal aberrations were also observed which indicates the genotoxicity of the extracts. All the extraction methods contributed significantly towards the drop in total MI. Compounds in the extracts is mitodepressive.     Makaluvamines are pyrroloiminoquinones isolated from Zyzzya sponges. Until now, they have been described as topoisomerase II inhibitors with cytotoxic effects in diverse tumor cell lines. In the present work, seven makaluvamines were tested in several antioxidant assays in primary cortical neurons and neuroblastoma cells. Among the alkaloids studied, makaluvamine J was the most active in all the assays. This compound was able to reduce the mitochondrial damage elicited by the well-known stressor H2O2. The antioxidant properties of makaluvamine J are related to an improvement of the endogenous antioxidant defenses of glutathione and catalase. SHSY5Y assays proved that this compound acts as a Nrf2 activator leading to an improvement of antioxidant defenses. A low concentration of 10 nM is able to reduce the reactive oxygen species release and maintain a correctmitochondrial function. Based on these results, non-substituted nitrogen in the pyrrole plus the presence of a p-hydroxystyryl without a double bond seems to be the most active structure with a complete antioxidant effect in neuronal cells.     Coastal and estuarine systems provide critical shark habitats due to their relatively high productivity and shallow, protected waters. The young (neonates, young‐of‐the‐year, and juveniles) of many coastal shark species occupy a diverse range of habitats and areas where they experience environmental variability, including acute and seasonal shifts in local salinities and temperatures. Although the location and functioning of essential shark habitats has been a focus in recent shark research, there is a paucity of data from the South Pacific. In this study, we document the temporal and spatial distribution, age class composition, and environmental parameters of young bull sharks (Carcharhinus leucas) in the Rewa, Sigatoka, and Navua Rivers, Fiji's three largest riverine systems. One hundred and seventy‐two young bull sharks were captured in fisheries‐independent surveys from January 2016 to April 2018. The vast majority of the captures were neonates. Seasonality in patterns of occurrence of neonate individuals suggests a defined parturition period during summer. Environmental parameters between the Rewa and the Sigatoka River differed significantly, as did the recorded young bull sharks abundance. According to the surveys, young bull sharks occur in all three rivers with the Rewa River likely representing essential habitat for newly born bull sharks. These results enhance the understanding of bull shark ecology in Fiji and provide a scientific basis for the implementation of local conservation strategies that contribute to the protection of critical habitats.     Nearshore deterioration of water quality in Pacific coastal waters is a growing problem, associated with increasing urban and industrial sewage discharges, and agricultural runoff. Published water quality studies in the Pacific region are limited in both number and scope, making it difficult to resolve the extent of the issue or quantify the variability of water quality across Pacific islands and countries. This study collected water quality measurements over three years in the coastal waters around the Island of Efate (Vanuatu) with majority of work carried out in Port Vila, its capital. Port Vila is the key urban centre for Vanuatu where the increasing population and pollution inputs are placing substantial pressure on the coastal environment. Highest concentrations of dissolved nutrients and suspended sediments were measured adjacent or near the urban drains that enter the coastal areas along the capital's seafront, highlighting many of the issues around anthropogenic inputs are linked to the increasing urbanisation in Port Vila Bay. We provide baseline data that explores variability of coastal water quality and these types of datasets for Pacific islands are a first step towards facilitating development of long-term monitoring programmes and informing coastal zone management decision making.

    The 2030 Agenda for Sustainable Development sets out a transformative vision which has yet to be realised for SDG14 and oceans. Recognition of the “indivisibility” of the Goals and enhanced integration of ocean governance support this transformation, but require at least multidisciplinary, or probably transdisciplinary, approaches. For regions which are highly dependent on development finance, a powerful leverage point for a transdisciplinary transformation is in the design of development investments. The work presented here identifies design features of ocean development-financed projects involving substantial amounts of research in two Pacific Small Island Developing States (SIDS): Fiji and the Solomon Islands. Transdisciplinary approaches were closely aligned to what is established as Mode 2 research modalities which focus on participation and multi-knowledge systems, as opposed to Mode 1 which have a predominantly scientific basis. From the literature an analytical indicator framework was developed which scored projects on their Modes of research within four categories: Product, Process, Policy and People. This framework was applied to five development-financed projects, and permitted the balance of Mode 1 and Mode 2 to be assessed and significant differences between projects identified. The work surfaces project features which can be embedded in the design of ocean investments which promote transdisciplinarity. This tractable and practical recognition of transdisciplinarity has connotations to the UN Ocean Decade in its ability to deliver on its transformation rhetoric. With capacity weaknesses and constrained financial resources in developing countries, and urgent ocean-related challenges especially in SIDS, moving to designed-in transdisciplinary and transformational outcomes remains a priority.

    AbstractSelective antibiotic treatment of white syndrome (WS)‐affected corals (Acropora muricata) from Fiji was used to identify 3 potential bacterial pathogens of the disease. Interestingly, the suite of bacterial associates of the disease was different to that recently identified using identical primer sets for WS on the GBR and in the Solomon Islands. In addition to the three bacterial pathogenic candidates and as previously shown for WS and more recently for white band disease (WBD) in the Caribbean, all samples of the disease were specifically associated with the histophagous ciliate Philaster lucinda. From the pattern of disease progression and histopathology in relation to the selective elimination of microbial groups, we conclude that these ‘white’ diseases are a result of a nonspecific bacterial infection and a ‘secondary’ infection by the P. lucinda ciliate. Although we have not observed the initiation of infection, a nonspecific, multispecies bacterial infection appears to be a corequirement for WS lesion progression and we hypothesize that the bacterial infection occurs initially, weakening the defences of the host to predation by the ciliates. Such ciliate histophagy gives rise to the characteristic white band of denuded coral skeleton that gives these diseases their names. The characteristics of the microbial communities of WBD and WS appear identical, and since the bacterial associates of WS vary geographically (and/or temporally), there appears to be no logical distinction between WS in the Indo‐Pacific and WBD in the Caribbean.

    Long-distance (>40-km) dispersal from marine reserves is poorly documented; yet, it can provide essential benefits such as seeding fished areas or connecting marine reserves into networks. From a meta-analysis, we suggest that the spatial scale of marine connectivity is underestimated due to the limited geographic extent of sampling designs. We also found that the largest marine reserves (>1000 km 2 ) are the most isolated. These findings have important implications for the assessment of evolutionary, ecological, and socio-economic long-distance benefits of marine reserves. We conclude that existing methods to infer dispersal should consider the up-to-date genomic advances and also expand the spatial scale of sampling designs. Incorporating long-distance connectivity in conservation planning will contribute to increase the benefits of marine reserve networks. International audience

    Water Quality issues in many Pacific countries are rising, with the increase in coastal populations and associated urban runoff but management requires contamination issues in the aquatic environment to be identified and prioritised. In Vanuatu and Solomon Islands there are few laboratories and resources to assess for the presence or impact of complex chemical contaminants. The extent and impact of chemical contamination of the marine and coastal environment is poorly described. Passive chemical samplers were used to measure a range of aquatic pollutants around the capital cities, Honiara (Solomon Islands) and Port Vila (Vanuatu). We detected a range of chemicals indicative of agricultural and industrial contamination and a few sites had concerning concentrations of specific hydrocarbons and pesticides. The rapid ecotoxicology test, Microtox, indicated toxic impacts in rivers, coastal sites and urban drains This work provides new data on chemical contamination and possible impacts of that contamination for both countries. The techniques could be applied widely across the region to generate critical data for environmental management, guide monitoring efforts and measure the impact of policy or land-use changes.

    Abstract In the Southwest Pacific Ocean, waters transit from the subtropical gyre before being redistributed equatorward and poleward. While the mean pathways are known, the contribution to the mixing and transport of the water from mesoscale eddies has not been comprehensively investigated. In this research, satellite altimetry data, combined with an eddy detection and tracking algorithm is used to investigate the distribution and surface characteristics of mesoscale eddies in this region of complex bathymetry (10°S–30°S, 140°E–190°E). Detected eddies are then colocalized with in situ data from Argo floats to determine their vertical structure and the effect of eddies on the water masses. The numerous islands affect the eddy behavior as most eddies are formed in the lee of islands, propagate westward and decay when encountering shallow bathymetry. Eddies are sparse and short‐lived in the tropical area north of Fiji, impacting only the top 200 meters of water. They do not appear to be able to trap and transport waters in this region. In the Coral Sea, a region of lateral shear between currents transporting waters of different origins, eddies are more numerous and energetic. They affect the water properties down to at least 500 m depth, and anticyclonic eddies trap water to ∼200 m, contributing to the upper thermocline waters mixing and transport. South of New Caledonia, mesoscale eddies are ubiquitous, with typical lifetimes longer than 5 months. They affect the temperature, salinity, and velocities down to ∼1,000 m depth, and weakly contribute to the mixing of lower thermocline waters. International audience

    Heavy rain events alter the biogeochemical outflows, affects water quality and ecosystem health within the coastal waters of small Pacific Islands. We characterized snapshots of the optical fingerprints of dissolved organic matter (DOM) sources together with the select nutrients, biogeochemical and physical variables for 10 stations in December 2017 in Laucala Bay, Fiji Islands. DOM absorption coefficients and fluorescence components were determined via spectrofluorometry and Parallel Factor Analysis identifying four components: Type M, two terrestrial (humic, fulvic) components, and a protein component linked to marine biological activity. Associations of DOM together with climate variables and the other tested variables were determined via principal component, hierarchical cluster, and cross-correlation (Pearson) analysis. All component s (together with most tested variables) displayed higher values (plumes) at the southwest coast consistent with surface currents outflow during the wet season. Type M component associated with two allochthonous fluorescent components signaling anthropogenic forcings via riverine outflows. Terrigenous inputs association with autochthonous chromophoric dissolved organic matter (CDOM) is indicative of tidal mixing, dilution, and bottom resuspension processes. Positive correlations of dissolved organic carbon (DOC) with nutrients (NOx, PO4) elucidates DOM being utilized as energy sources. The positive correlation of DON with nutrients (NOx, PO4, Si(OH)4) reflects the role of DON as a nutrient source consistent with chlorophyll plume formation International audience

    There has been a growing interest in medicinal plants by the locals of the Pacific Islands since it is readily available. Annona muricata, commonly called soursop is one of such plants which is popular because it is being used to treat initial stages of cancer due to its antioxidant properties. This study investigated various extraction methods which could be used to compare and see the effectiveness of each on the chromosomes. The cytotoxicity and genotoxicity effect were studied using Allium cepa root tip cells. Eight onion bulbs were subjected to the treatment groups which were made using methanol, water and hexane extracts of A. muricata. Three different concentrations from each extraction solvent were used. The A. cepa root tip cells were subjected for 24 h to three different leaf extractions after left to grow in water. Concentrations of 12.5, 25 and 50 g/L were used. After the treatment, onion bulbs were put back in water to recover. Hence, 5 root tips from each bulb were removed (before treatment, treatment and recovery) with cells undergoing mitosis counted under 400x of a compound microscope. All studied concentrations of A. muricata methanol extract showed significant (p≤0.05) drop in the mean mitotic index when compared with control (mean MI reduced by 9.9, 44.4 and 54.2% for 12.5, 25 and 50 g/L respectively). A. muricata water extract also showed significant differences at 12.5 g/L where mitotic index (MI) was reduced by 8.6%. At 25 g/L, the reduction was by 35.3% and by 42.8% at 50 g/L. Drastic effect (p≤0.05) was observed with the soursop-hexane extract that reduced the MI significantly in 12.5 g/L by 5.0% and by 29.7% at 25 g/L. A huge reduction in MI was observed at 50 g/L where MI declined by 72.8%. Chromosomal aberrations were also observed which indicates the genotoxicity of the extracts. All the extraction methods contributed significantly towards the drop in total MI. Compounds in the extracts is mitodepressive.     Makaluvamines are pyrroloiminoquinones isolated from Zyzzya sponges. Until now, they have been described as topoisomerase II inhibitors with cytotoxic effects in diverse tumor cell lines. In the present work, seven makaluvamines were tested in several antioxidant assays in primary cortical neurons and neuroblastoma cells. Among the alkaloids studied, makaluvamine J was the most active in all the assays. This compound was able to reduce the mitochondrial damage elicited by the well-known stressor H2O2. The antioxidant properties of makaluvamine J are related to an improvement of the endogenous antioxidant defenses of glutathione and catalase. SHSY5Y assays proved that this compound acts as a Nrf2 activator leading to an improvement of antioxidant defenses. A low concentration of 10 nM is able to reduce the reactive oxygen species release and maintain a correctmitochondrial function. Based on these results, non-substituted nitrogen in the pyrrole plus the presence of a p-hydroxystyryl without a double bond seems to be the most active structure with a complete antioxidant effect in neuronal cells.     Coastal and estuarine systems provide critical shark habitats due to their relatively high productivity and shallow, protected waters. The young (neonates, young‐of‐the‐year, and juveniles) of many coastal shark species occupy a diverse range of habitats and areas where they experience environmental variability, including acute and seasonal shifts in local salinities and temperatures. Although the location and functioning of essential shark habitats has been a focus in recent shark research, there is a paucity of data from the South Pacific. In this study, we document the temporal and spatial distribution, age class composition, and environmental parameters of young bull sharks (Carcharhinus leucas) in the Rewa, Sigatoka, and Navua Rivers, Fiji's three largest riverine systems. One hundred and seventy‐two young bull sharks were captured in fisheries‐independent surveys from January 2016 to April 2018. The vast majority of the captures were neonates. Seasonality in patterns of occurrence of neonate individuals suggests a defined parturition period during summer. Environmental parameters between the Rewa and the Sigatoka River differed significantly, as did the recorded young bull sharks abundance. According to the surveys, young bull sharks occur in all three rivers with the Rewa River likely representing essential habitat for newly born bull sharks. These results enhance the understanding of bull shark ecology in Fiji and provide a scientific basis for the implementation of local conservation strategies that contribute to the protection of critical habitats.     Nearshore deterioration of water quality in Pacific coastal waters is a growing problem, associated with increasing urban and industrial sewage discharges, and agricultural runoff. Published water quality studies in the Pacific region are limited in both number and scope, making it difficult to resolve the extent of the issue or quantify the variability of water quality across Pacific islands and countries. This study collected water quality measurements over three years in the coastal waters around the Island of Efate (Vanuatu) with majority of work carried out in Port Vila, its capital. Port Vila is the key urban centre for Vanuatu where the increasing population and pollution inputs are placing substantial pressure on the coastal environment. Highest concentrations of dissolved nutrients and suspended sediments were measured adjacent or near the urban drains that enter the coastal areas along the capital's seafront, highlighting many of the issues around anthropogenic inputs are linked to the increasing urbanisation in Port Vila Bay. We provide baseline data that explores variability of coastal water quality and these types of datasets for Pacific islands are a first step towards facilitating development of long-term monitoring programmes and informing coastal zone management decision making.

    The 2030 Agenda for Sustainable Development sets out a transformative vision which has yet to be realised for SDG14 and oceans. Recognition of the “indivisibility” of the Goals and enhanced integration of ocean governance support this transformation, but require at least multidisciplinary, or probably transdisciplinary, approaches. For regions which are highly dependent on development finance, a powerful leverage point for a transdisciplinary transformation is in the design of development investments. The work presented here identifies design features of ocean development-financed projects involving substantial amounts of research in two Pacific Small Island Developing States (SIDS): Fiji and the Solomon Islands. Transdisciplinary approaches were closely aligned to what is established as Mode 2 research modalities which focus on participation and multi-knowledge systems, as opposed to Mode 1 which have a predominantly scientific basis. From the literature an analytical indicator framework was developed which scored projects on their Modes of research within four categories: Product, Process, Policy and People. This framework was applied to five development-financed projects, and permitted the balance of Mode 1 and Mode 2 to be assessed and significant differences between projects identified. The work surfaces project features which can be embedded in the design of ocean investments which promote transdisciplinarity. This tractable and practical recognition of transdisciplinarity has connotations to the UN Ocean Decade in its ability to deliver on its transformation rhetoric. With capacity weaknesses and constrained financial resources in developing countries, and urgent ocean-related challenges especially in SIDS, moving to designed-in transdisciplinary and transformational outcomes remains a priority.

    AbstractSelective antibiotic treatment of white syndrome (WS)‐affected corals (Acropora muricata) from Fiji was used to identify 3 potential bacterial pathogens of the disease. Interestingly, the suite of bacterial associates of the disease was different to that recently identified using identical primer sets for WS on the GBR and in the Solomon Islands. In addition to the three bacterial pathogenic candidates and as previously shown for WS and more recently for white band disease (WBD) in the Caribbean, all samples of the disease were specifically associated with the histophagous ciliate Philaster lucinda. From the pattern of disease progression and histopathology in relation to the selective elimination of microbial groups, we conclude that these ‘white’ diseases are a result of a nonspecific bacterial infection and a ‘secondary’ infection by the P. lucinda ciliate. Although we have not observed the initiation of infection, a nonspecific, multispecies bacterial infection appears to be a corequirement for WS lesion progression and we hypothesize that the bacterial infection occurs initially, weakening the defences of the host to predation by the ciliates. Such ciliate histophagy gives rise to the characteristic white band of denuded coral skeleton that gives these diseases their names. The characteristics of the microbial communities of WBD and WS appear identical, and since the bacterial associates of WS vary geographically (and/or temporally), there appears to be no logical distinction between WS in the Indo‐Pacific and WBD in the Caribbean.

    Long-distance (>40-km) dispersal from marine reserves is poorly documented; yet, it can provide essential benefits such as seeding fished areas or connecting marine reserves into networks. From a meta-analysis, we suggest that the spatial scale of marine connectivity is underestimated due to the limited geographic extent of sampling designs. We also found that the largest marine reserves (>1000 km 2 ) are the most isolated. These findings have important implications for the assessment of evolutionary, ecological, and socio-economic long-distance benefits of marine reserves. We conclude that existing methods to infer dispersal should consider the up-to-date genomic advances and also expand the spatial scale of sampling designs. Incorporating long-distance connectivity in conservation planning will contribute to increase the benefits of marine reserve networks. International audience
