We have applied to these images an iterative multi-scale wavelet analysis and reconstruction technique, which allows to subtract stars and galaxies from the original images. We detect a zone of diffuse emission south west of the cluster center (DS1), and a second faint object (ComDif), within DS1. Another diffuse source (DS2) may be detected, at lower confidence level, north east of the center. These sources of diffuse light contribute to 10-15% of the total visible light in the cluster. Whether they are independent entities or are part of the very elliptical external envelope of the central galaxy remains unclear. VLT VIMOS integral field spectroscopy reveals a faint continuum at the positions of DS1 and ComDif but do not allow to compute a redshift. A hierarchical substructure detection method reveals the presence of several galaxy pairs and groups defining a similar direction as the one drawn by the DS1-central galaxy-DS2 axis. The analysis of archive XMM-Newton and Chandra observations shows X-ray emission elongated in the same direction. The X-ray temperature map shows the presence of a cool core, a broad cool zone stretching from north to south and hotter regions towards the north east, south west and north west. This possibly suggests shock fronts along these directions produced by infalling material. These various data are consistent with a picture in which diffuse sources are concentrations of tidal debris and harassed matter expelled from infalling galaxies by tidal stripping and undergoing an accretion process onto the central cluster galaxy; as such, they are expected to be found along the main infall directions. Manzoni;International audience; Cannabis exposure during the perinatal period results in varied and significant consequences in affected offspring. The prevalence of detrimental outcomes of perinatal cannabis exposure is likely to increase in tandem with the broadening of legalization and acceptance of the drug. As such, it is crucial to highlight the immediate and protracted consequences of cannabis exposure on pre-and post-natal development. Here, we identify lasting changes in neurons' learning flexibility (synaptic plasticity) and epigenetic misregulation in animal models of perinatal cannabinoid exposure (using synthetic cannabinoids or active components of the cannabis plant) in addition to significant alterations in social behavior and executive functions. These findings are supported by epidemiological data indicating similar behavioral outcomes throughout life in human offspring exposed to cannabis during pregnancy. Further, we indicate important lingering questions regarding accurate modeling of perinatal cannabis exposure as well as the need for sex-and agedependent outcome measures in future studies.

The description of these cases was published in the Transactions of the American Neurological Association ,1 ,2 and led in the following year to what is considered the first comprehensive description of progressive supranuclear palsy (PSP) published.3 Here, Richardson and coworkers described the clinical features in nine patients and detailed the neuropathological changes in seven of them. In this report, the authors fended off arguments that the condition in the patients they described reflected a genetic mutation or environmental toxin geographically confined to the Toronto area. Morace; CANTORE, Giampaolo; MIRABELLA, GIOVANNI;In humans, the ability to withhold manual motor responses seems to rely on a right-lateralized frontal–basal ganglia–thalamic network, including the pre-supplementary motor area and the inferior frontal gyrus (IFG). These areas should drive subthalamic nuclei to implement movement inhibition via the hyperdirect pathway. The output of this network is expected to influence those cortical areas underlying limb movement preparation and initiation, i.e., premotor (PMA) and primary motor (M1) cortices. Electroencephalographic (EEG) studies have shown an enhancement of the N200/P300 complex in the event-related potentials (ERPs) when a planned reaching movement is successfully stopped after the presentation of an infrequent stop-signal. PMA and M1 have been suggested as possible neural sources of this ERP complex but, due to the limited spatial resolution of scalp EEG, it is not yet clear which cortical areas contribute to its generation. To elucidate the role of motor cortices, we recorded epicortical ERPs from the lateral surface of the fronto-temporal lobes of five pharmacoresistant epileptic patients performing a reaching version of the countermanding task while undergoing presurgical monitoring. We consistently found a stereotyped ERP complex on a single-trial level when a movement was successfully cancelled. These ERPs were selectively expressed in M1, PMA, and Brodmann's area (BA) 9 and their onsets preceded the end of the stop process, suggesting a causal involvement in this executive function. Such ERPs also occurred in unsuccessful-stop (US) trials, that is, when subjects moved despite the occurrence of a stop-signal, mostly when they had long reaction times (RTs). These findings support the hypothesis that motor cortices are the final target of the inhibitory command elaborated by the frontal–basal ganglia–thalamic network.

In this context, the purpose of the present study was to investigate possible protective effects of RA on mismatch negativity (MMN) component of auditory event-related potentials (AERPs) as an indicator of auditory discrimination and echoic memory in the ovariectomized (OVX) rats injected with D-galactose combined with neurochemical and histological analyses. Ninety female Wistar rats were randomly divided into six groups: sham control (5); RA-treated (R); OVX (O); OVX + RA-treated (OR); OVX+ D-galactose-treated (OD); OVX+ D-galactose + RA treated (ODR). Eight weeks later, MMN responses were recorded using the oddball condition. An amplitude reduction of some components of AERPs was observed due to ovariectomy with or without D-galactose administiration and these reduction patterns were diverse for different electrode locations. MMN amplitudes were significantly lower over temporal and right frontal locations in the o and OD groups versus the S and R groups, which was accompanied by increased thiobarbituric acid reactive substances (TBARS) and hydroxy-2-nonenal (4-HNE) levels. RA treatment significantly increased AERP/MMN amplitudes and lowered the TBARS/4-HNE levels in the OR and ODR groups versus the 0 and OD groups, respectively. Our findings support the potential benefit of RA in the prevention of auditory distortion related to the estrogen deficiency and D-galactose administration at least partly by antioxidant actions. In particular, we compared the effects of a continuous LN stimulation at low-frequency (tonic stimulation) with those induced by high frequency pulse trains (phasic stimulation). The observed results show that the stimulation of LN induces c-fos expression in a significant proportion of neurones in the contralateral AON. Phasic stimulation was slightly more effective than tonic stimulation in producing c-fos expression.

P.; Verderio C.; Fumagalli M.;Contrasting myelin damage through the generation of new myelinating oligodendrocytes represents a promising approach to promote functional recovery after stroke. Here, we asked whether activation of microglia and monocyte-derived macrophages affects the regenerative process sustained by G protein-coupled receptor 17 (GPR17)-expressing oligodendrocyte precursor cells (OPCs), a subpopulation of OPCs specifically reacting to ischemic injury. GPR17-iCreER(T2):CAG-eGFP reporter mice were employed to trace the fate of GPR17-expressing OPCs, labeled by the green fluorescent protein (GFP), after permanent middle cerebral artery occlusion. By microglia/macrophages pharmacological depletion studies, we show that innate immune cells favor GFP(+) OPC reaction and limit myelin damage early after injury, whereas they lose their pro-resolving capacity and acquire a dystrophic “senescent-like” phenotype at later stages. Intracerebral infusion of regenerative microglia-derived extracellular vesicles (EVs) restores protective microglia/macrophages functions, limiting their senescence during the post-stroke phase, and enhances the maturation of GFP(+) OPCs at lesion borders, resulting in ameliorated neurological functionality. In vitro experiments show that EV-carried transmembrane tumor necrosis factor (tmTNF) mediates the pro-differentiating effects on OPCs, with future implications for regenerative therapies.

Obstructive sleep apnea (OSA) is commonly associated with neurocognitive impairments that have not been consistently related to specific brain structure abnormalities. Knowledge of the brain structures involved in OSA and the corresponding functional implications could provide clues to the pathogenesis of cognitive impairment and its reversibility in this disorder.. OBJECTIVES:. . To investigate the cognitive deficits and the corresponding brain morphology changes in OSA, and the modifications after treatment, using combined neuropsychologic testing and voxel-based morphometry.. METHODS:. . A total of 17 patients treatment-naive to sleep apnea and 15 age-matched healthy control subjects underwent a sleep study, cognitive tests, and magnetic resonance imaging. After 3 months of treatment, cognitive and imaging data were collected to assess therapy efficacy.. MEASUREMENTS AND MAIN RESULTS:. . Neuropsychologic results in pretreatment OSA showed impairments in most cognitive areas, and in mood and sleepiness. These impairments were associated with focal reductions of gray-matter volume in the left hippocampus (entorhinal cortex), left posterior parietal cortex, and right superior frontal gyrus. After treatment, we observed significant improvements involving memory, attention, and executive-functioning that paralleled gray-matter volume increases in hippocampal and frontal structures.. CONCLUSIONS:. . The cognitive and structural deficits in OSA may be secondary to sleep deprivation and repetitive nocturnal intermittent hypoxemia. These negative effects may be recovered by consistent and thorough treatment. Our findings highlight the importance of early diagnosis and successful treatment of this disorder."

The idea that elementary processors move within a network to get a proper position is borne out both by biological neurons in the brain morphogenesis and by agents within social networks. From the former, we take inspiration to devise a medium-term project for new artificial neural network training procedures where mobile neurons exchange data only when they are close to one another in a proper space (are in contact). From the latter, we accumulate mobility tracks experience. We focus on the preliminary step of characterizing the elapsed time between neuron contacts, which results from a spatial process fitting in the family of random processes with memory, where chasing neurons are stochastically driven by the goal of hitting target neurons. Thus, we add an unprecedented mobility model to the literature in the field, introducing a distribution law of the intercontact times that merges features of both negative exponential and Pareto distribution laws. We give a constructive description and implementation of our model, as well as a short analytical form whose parameters are suitably estimated in terms of confidence intervals from experimental data. Numerical experiments show the model and related inference tools to be sufficiently robust to cope with two main requisites for its exploitation in a neural network: the nonindependence of the observed intercontact times and the feasibility of the model inversion problem to infer suitable mobility parameters.

We have applied to these images an iterative multi-scale wavelet analysis and reconstruction technique, which allows to subtract stars and galaxies from the original images. We detect a zone of diffuse emission south west of the cluster center (DS1), and a second faint object (ComDif), within DS1. Another diffuse source (DS2) may be detected, at lower confidence level, north east of the center. These sources of diffuse light contribute to 10-15% of the total visible light in the cluster. Whether they are independent entities or are part of the very elliptical external envelope of the central galaxy remains unclear. VLT VIMOS integral field spectroscopy reveals a faint continuum at the positions of DS1 and ComDif but do not allow to compute a redshift. A hierarchical substructure detection method reveals the presence of several galaxy pairs and groups defining a similar direction as the one drawn by the DS1-central galaxy-DS2 axis. The analysis of archive XMM-Newton and Chandra observations shows X-ray emission elongated in the same direction. The X-ray temperature map shows the presence of a cool core, a broad cool zone stretching from north to south and hotter regions towards the north east, south west and north west. This possibly suggests shock fronts along these directions produced by infalling material. These various data are consistent with a picture in which diffuse sources are concentrations of tidal debris and harassed matter expelled from infalling galaxies by tidal stripping and undergoing an accretion process onto the central cluster galaxy; as such, they are expected to be found along the main infall directions. Manzoni;International audience; Cannabis exposure during the perinatal period results in varied and significant consequences in affected offspring. The prevalence of detrimental outcomes of perinatal cannabis exposure is likely to increase in tandem with the broadening of legalization and acceptance of the drug. As such, it is crucial to highlight the immediate and protracted consequences of cannabis exposure on pre-and post-natal development. Here, we identify lasting changes in neurons' learning flexibility (synaptic plasticity) and epigenetic misregulation in animal models of perinatal cannabinoid exposure (using synthetic cannabinoids or active components of the cannabis plant) in addition to significant alterations in social behavior and executive functions. These findings are supported by epidemiological data indicating similar behavioral outcomes throughout life in human offspring exposed to cannabis during pregnancy. Further, we indicate important lingering questions regarding accurate modeling of perinatal cannabis exposure as well as the need for sex-and agedependent outcome measures in future studies.

The description of these cases was published in the Transactions of the American Neurological Association ,1 ,2 and led in the following year to what is considered the first comprehensive description of progressive supranuclear palsy (PSP) published.3 Here, Richardson and coworkers described the clinical features in nine patients and detailed the neuropathological changes in seven of them. In this report, the authors fended off arguments that the condition in the patients they described reflected a genetic mutation or environmental toxin geographically confined to the Toronto area. Morace; CANTORE, Giampaolo; MIRABELLA, GIOVANNI;In humans, the ability to withhold manual motor responses seems to rely on a right-lateralized frontal–basal ganglia–thalamic network, including the pre-supplementary motor area and the inferior frontal gyrus (IFG). These areas should drive subthalamic nuclei to implement movement inhibition via the hyperdirect pathway. The output of this network is expected to influence those cortical areas underlying limb movement preparation and initiation, i.e., premotor (PMA) and primary motor (M1) cortices. Electroencephalographic (EEG) studies have shown an enhancement of the N200/P300 complex in the event-related potentials (ERPs) when a planned reaching movement is successfully stopped after the presentation of an infrequent stop-signal. PMA and M1 have been suggested as possible neural sources of this ERP complex but, due to the limited spatial resolution of scalp EEG, it is not yet clear which cortical areas contribute to its generation. To elucidate the role of motor cortices, we recorded epicortical ERPs from the lateral surface of the fronto-temporal lobes of five pharmacoresistant epileptic patients performing a reaching version of the countermanding task while undergoing presurgical monitoring. We consistently found a stereotyped ERP complex on a single-trial level when a movement was successfully cancelled. These ERPs were selectively expressed in M1, PMA, and Brodmann's area (BA) 9 and their onsets preceded the end of the stop process, suggesting a causal involvement in this executive function. Such ERPs also occurred in unsuccessful-stop (US) trials, that is, when subjects moved despite the occurrence of a stop-signal, mostly when they had long reaction times (RTs). These findings support the hypothesis that motor cortices are the final target of the inhibitory command elaborated by the frontal–basal ganglia–thalamic network.

D.; Olgar Y.; Yargicoglu P.;Rosmarinic acid (RA), which has multiple bioactive properties, might be a useful agent for protecting central nervous system against age related alterations. In this context, the purpose of the present study was to investigate possible protective effects of RA on mismatch negativity (MMN) component of auditory event-related potentials (AERPs) as an indicator of auditory discrimination and echoic memory in the ovariectomized (OVX) rats injected with D-galactose combined with neurochemical and histological analyses. Ninety female Wistar rats were randomly divided into six groups: sham control (5); RA-treated (R); OVX (O); OVX + RA-treated (OR); OVX+ D-galactose-treated (OD); OVX+ D-galactose + RA treated (ODR). Eight weeks later, MMN responses were recorded using the oddball condition. An amplitude reduction of some components of AERPs was observed due to ovariectomy with or without D-galactose administiration and these reduction patterns were diverse for different electrode locations. MMN amplitudes were significantly lower over temporal and right frontal locations in the o and OD groups versus the S and R groups, which was accompanied by increased thiobarbituric acid reactive substances (TBARS) and hydroxy-2-nonenal (4-HNE) levels. RA treatment significantly increased AERP/MMN amplitudes and lowered the TBARS/4-HNE levels in the OR and ODR groups versus the 0 and OD groups, respectively. Our findings support the potential benefit of RA in the prevention of auditory distortion related to the estrogen deficiency and D-galactose administration at least partly by antioxidant actions. In particular, we compared the effects of a continuous LN stimulation at low-frequency (tonic stimulation) with those induced by high frequency pulse trains (phasic stimulation). The observed results show that the stimulation of LN induces c-fos expression in a significant proportion of neurones in the contralateral AON. Phasic stimulation was slightly more effective than tonic stimulation in producing c-fos expression.

P.; Verderio C.; Fumagalli M.;Contrasting myelin damage through the generation of new myelinating oligodendrocytes represents a promising approach to promote functional recovery after stroke. Here, we asked whether activation of microglia and monocyte-derived macrophages affects the regenerative process sustained by G protein-coupled receptor 17 (GPR17)-expressing oligodendrocyte precursor cells (OPCs), a subpopulation of OPCs specifically reacting to ischemic injury. GPR17-iCreER(T2):CAG-eGFP reporter mice were employed to trace the fate of GPR17-expressing OPCs, labeled by the green fluorescent protein (GFP), after permanent middle cerebral artery occlusion. By microglia/macrophages pharmacological depletion studies, we show that innate immune cells favor GFP(+) OPC reaction and limit myelin damage early after injury, whereas they lose their pro-resolving capacity and acquire a dystrophic “senescent-like” phenotype at later stages. Intracerebral infusion of regenerative microglia-derived extracellular vesicles (EVs) restores protective microglia/macrophages functions, limiting their senescence during the post-stroke phase, and enhances the maturation of GFP(+) OPCs at lesion borders, resulting in ameliorated neurological functionality. In vitro experiments show that EV-carried transmembrane tumor necrosis factor (tmTNF) mediates the pro-differentiating effects on OPCs, with future implications for regenerative therapies.

Obstructive sleep apnea (OSA) is commonly associated with neurocognitive impairments that have not been consistently related to specific brain structure abnormalities. Knowledge of the brain structures involved in OSA and the corresponding functional implications could provide clues to the pathogenesis of cognitive impairment and its reversibility in this disorder.. OBJECTIVES:. . To investigate the cognitive deficits and the corresponding brain morphology changes in OSA, and the modifications after treatment, using combined neuropsychologic testing and voxel-based morphometry.. METHODS:. . A total of 17 patients treatment-naive to sleep apnea and 15 age-matched healthy control subjects underwent a sleep study, cognitive tests, and magnetic resonance imaging. After 3 months of treatment, cognitive and imaging data were collected to assess therapy efficacy.. MEASUREMENTS AND MAIN RESULTS:. . Neuropsychologic results in pretreatment OSA showed impairments in most cognitive areas, and in mood and sleepiness. These impairments were associated with focal reductions of gray-matter volume in the left hippocampus (entorhinal cortex), left posterior parietal cortex, and right superior frontal gyrus. After treatment, we observed significant improvements involving memory, attention, and executive-functioning that paralleled gray-matter volume increases in hippocampal and frontal structures.. CONCLUSIONS:. . The cognitive and structural deficits in OSA may be secondary to sleep deprivation and repetitive nocturnal intermittent hypoxemia. These negative effects may be recovered by consistent and thorough treatment. Our findings highlight the importance of early diagnosis and successful treatment of this disorder."

The idea that elementary processors move within a network to get a proper position is borne out both by biological neurons in the brain morphogenesis and by agents within social networks. From the former, we take inspiration to devise a medium-term project for new artificial neural network training procedures where mobile neurons exchange data only when they are close to one another in a proper space (are in contact). From the latter, we accumulate mobility tracks experience. We focus on the preliminary step of characterizing the elapsed time between neuron contacts, which results from a spatial process fitting in the family of random processes with memory, where chasing neurons are stochastically driven by the goal of hitting target neurons. Thus, we add an unprecedented mobility model to the literature in the field, introducing a distribution law of the intercontact times that merges features of both negative exponential and Pareto distribution laws. We give a constructive description and implementation of our model, as well as a short analytical form whose parameters are suitably estimated in terms of confidence intervals from experimental data. Numerical experiments show the model and related inference tools to be sufficiently robust to cope with two main requisites for its exploitation in a neural network: the nonindependence of the observed intercontact times and the feasibility of the model inversion problem to infer suitable mobility parameters.
