A large proportion of European biodiversity today depends on habitat provided by low‐intensity farming practices, yet this resource is declining as European agriculture intensifies. Within the European Union, particularly the central and eastern new member states have retained relatively large areas of species‐rich farmland, but despite increased investment in nature conservation here in recent years, farmland biodiversity trends appear to be worsening. Although the high biodiversity value of Central and Eastern European farmland has long been reported, the amount of research in the international literature focused on farmland biodiversity in this region remains comparatively tiny, and measures within the EU Common Agricultural Policy are relatively poorly adapted to support it. In this opinion study, we argue that, 10 years after the accession of the first eastern EU new member states, the continued under‐representation of the low‐intensity farmland in Central and Eastern Europe in the international literature and EU policy is impeding the development of sound, evidence‐based conservation interventions. The biodiversity benefits for Europe of existing low‐intensity farmland, particularly in the central and eastern states, should be harnessed before they are lost. Instead of waiting for species‐rich farmland to further decline, targeted research and monitoring to create locally appropriate conservation strategies for these habitats is needed now. Their continued existence depends on some form of human activity, for either production or conservation purposes, or both. We examined the share of semi-natural grasslands within the general grassland areas in boreal Europe. We reviewed research literature across the region to compile evidence on semi-natural grasslands and other semi-natural habitats, such as wooded pastures, in respect to a range of topics such as ecology, land-use change, socioeconomics, and production. We also explored drivers of the research agenda and outlined future research needs. Challenges are faced when defining and quantifying semi-natural habitats even across a restricted region. Agricultural development and other policies clearly impact the research agenda in various countries. There are recent signs of a shift from classical ecological studies toward more multidisciplinary and integrated research. To sufficiently address the threats faced by semi-natural habitats, political and research frameworks in the European Union should pay more attention to the social-ecological complexity inherent in their management and should support the engagement of various actors into participatory governance processes. This is in line with a full-farm approach implicit in high nature value farming systems. Peer reviewed

Backgroun Ecosystem services (ES) mapping is becoming mainstream in many sustainability assessments, but its impact on real world decision-making is still limited. Robustness, enduser relevance and transparency have been identified as key attributes needed for effective ES mapping. However, these requirements are not always met due to multiple challenges, referred to here as bottlenecks, that scientists, practitioners, policy makers and users from other public and private sectors encounter along the mapping process. A selection of commonly encountered ES mapping bottlenecks that relate to seven themes: i) map-maker map-user interaction ii) nomenclature and ontologies iii) skills and background iv) data and maps availability v) methods-selection vi) technical difficulties and vii) over-simplification of mapping process/output. The authors synthesise the variety of solutions already applied by map-makers and map-users to mitigate or cope with these bottlenecks and discuss the emerging trade-offs amongst different solutions. Tackling the bottlenecks described here is a crucial first step towards more effective ES mapping, which can in turn ensure the adequate impact of ES mapping in decision-making. In MCDM, a crowd as a whole, or its individual members, generally make decisions free from any influence of valence, arousal, emotional state or environment. In contrast, various theories dealing with crowd psychology (Gustave Le Bon, Freudian, Deindividuation, Convergence, Emergent norm, Social identity) analyze, in one form or another, the emotions of the crowd. According to above theories, crowd is influenced by a range of behavioral factors, such as physical, social, psychological, culture, norms, and emotions. It can be argued that the emotional state, valence and arousal of crowds affect their decision making to a considerable degree and multiple criteria crowd behavior modeling must, therefore, consider this impact as well. In this light, the integration of crowd simulation and biometric methods, behavioral operations research and emotions in decision making has taken a prominent place as it leads to a better understanding of crowd emotions and crowd decision making. In this context, the authors developed the Affective Analytics of Demonstration Sites (ANDES) that added to this body of research in four ways. The crowd analysis and simulations conducted with ANDES used a neuro decision matrix. The matrix contains a detailed description of demonstration sites (public spaces) in question and the emotions, valence, arousal and physiological parameters of people present there. With ANDES’s Remote Sensor Network, emotional (emotions, valence, arousal) and physiological (average crowd facial temperature, crowd composition by gender and age group, etc.) parameters of people present at demonstration sites can be mapped. ANDES can assist experts in more effective implementations of public spaces planning and a participation process by attendees by collecting and examining various layers of data on the emotional and physiological parameters of visitors based on a visitors-centric public spaces planning approach. ANDES can determine the public space and real estate values.

Este ctenostomado ha sido considerado por mucho tiempo como una especie nativa del Mar Mediterráneo, pero se ha sugerido recientemente que es originario del Caribe. Es probable que haya sido introducido incrustado en los cascos de los barcos. The spaghetti bryozoan Amathia verticillata, formerly known as Zoobotryon verticillatum, was first described in 1822 from Naples, Italy, although this species was already present in 1807 at Cadiz, Spain. This ctenostome has long been considered a native species in the Mediterranean Sea but it has recently been suggested to be of Caribbean origin. It is most likely to have been introduced by vessels as hull fouling. This pseudo-indigenous species, i.e. a non-indigenous species (NIS) having been perceived to be native, has been found in several marinas and harbours within the Mediterranean Sea. In November 2014, this bryozoan species was abundant in the La Grande Motte marina on the south coast of France. Several thousand colonies were estimated to be present within this marina attached to the floating pontoon units that supported a floating boardwalk. Of the berthed craft examined, 31% were fouled with this species, and it was occasionally a prominent fouling species. Several macroinvertebrate species were associated with A. verticillata colonies, including some NIS, Paracerceis sculpta, Paranthura japonica and Caprella scaura, that are recorded for the first time from the Mediterranean coast of France. A. verticillata might support their transfer elsewhere by providing a habitat and substrate when attached to vessel hulls.

Due to practical reasons, the most suitable positions for modifications are C5 of pyrimidines and C7 of purines. Unfortunately, by using these two positions only, one cannot expand a repertoire of modified nucleotides to a maximum. Here, we demonstrate the synthesis and enzymatic incorporation of novel N-4-acylated 2'-deoxycytidine nucleotides (dC(Acyl)). We find that a variety of family A and B DNA polymerases efficiently use dC(Acyl)TPs as substrates. In addition to the formation of complementary C-Acyl center dot G pair, a strong basepairing between N-4-acyl-cytosine and adenine takes place when Taq, Klenow fragment (exo-), Bsm and KOD XL DNA polymerases are used for the primer extension reactions. In contrast, a proofreading phi29 DNA polymerase successfully utilizes dC(Acyl)TPs but is prone to form C-Acyl center dot A base pair under the same conditions. Moreover, we show that terminal deoxynucleotidyl transferase is able to incorporate as many as several hundred N-4-acylated-deoxycytidine nucleotides. These data reveal novel N-4-acylated deoxycytidine nucleotides as beneficial substrates for the enzymatic synthesis of modified DNA, which can be further applied for specific labelling of DNA fragments, selection of aptamers or photoimmobilization.

This work compares meteorological results from different regional climate model (RCM) implementations in the Mediterranean area, with a focus on the northern Adriatic Sea. The need to use these datasets as atmospheric forcings (wind and atmospheric pressure fields) for coastal hydrodynamic models to assess future changes in the coastal hydrodynamics, is the basis of the presented analysis. It would allow the assessment of uncertainties due to atmospheric forcings in providing coastal current, surge and wave climate changes from future implementations of hydrodynamic models. Two regional climate models, with different spatial resolutions, downscaled from two different global climate models (whose atmospheric components are, respectively, ECHAM4 and ECHAM5), were considered. In particular, the RCM delivered wind and atmospheric pressure fields were compared with measurements at four stations along the Italian Adriatic coast. The analyses were conducted using a past control period, 1960–1990, and the A1B IPCC future scenario (2070–2100). The chosen scenario corresponds to a world of very rapid economic and demographic growth that peaks in mid-century, with a rapid introduction of new efficient technologies, which balance fossil and non-fossil resources (IPCC, 2007). Consideration is given to the accuracy of each model at reproducing the basic statistics and the trends. The role of models' spatial resolution in reproducing global and local scale meteorological processes is also discussed. The Adriatic Sea climate is affected by the orography that produces a strengthening of north-eastern katabatic winds like bora. Therefore, spatial model resolution, both for orography and for a better resolution of coastline (Cavaleri et al., 2010), is one of the important factors in providing more realistic wind forcings for future hydrodynamic models implementations. However, also the characteristics in RCM setup and parameterization can explain differences between the datasets. The analysis from an ensemble of model implementation would provide more robust indications on climatic wind and atmospheric pressure variations. The scenario-control comparison shows a general increase in the mean atmospheric pressure values while a decrease in mean wind speed and in extreme wind events is seen, particularly for the datasets with higher spatial resolution.

Here, we aimed to describe the early-life exposome. Specifically, we focused on the correlations between multiple environmental exposures, their patterns and their variability across European regions and across time (pregnancy and childhood periods). We relied on the Human Early-Life Exposome (HELIX) project, in which 87 environmental exposures during pregnancy and 122 during the childhood period (grouped in 19 exposure groups) were assessed in 1301 pregnant mothers and their children at 6-11 years in 6 European birth cohorts. Some correlations between exposures in the same exposure group reached high values above 0.8. The median correlation within exposure groups was >0.3 for many exposure groups, reaching 0.69 for water disinfection by products in pregnancy and 0.67 for the meteorological group in childhood. Median correlations between different exposure groups rarely reached 0.3. Some correlations were driven by cohort-level associations (e.g. air pollution and chemicals). Ten principal components explained 45% and 39% of the total variance in the pregnancy and childhood exposome, respectively, while 65 and 90 components were required to explain 95% of the exposome variability. Correlations between maternal (pregnancy) and childhood exposures were high (>0.6) for most exposures modeled at the residential address (e.g. air pollution), but were much lower and even close to zero for some chemical exposures. In conclusion, the early life exposome was high dimensional, meaning that it cannot easily be measured by or reduced to fewer components. Correlations between exposures from different exposure groups were much lower than within exposure groups, which have important implications for co-exposure confounding in multiple exposure studies. Also, we observed the early life exposome to be variable over time and to vary by cohort, so measurements at one time point or one place will not capture its complexities. Many of the next generation of global climate models will include aerosol schemes which explicitly simulate the microphysical processes that determine the particle size distribution. These models enable aerosol optical properties and cloud condensation nuclei (CCN) concentrations to be determined by fundamental aerosol processes, which should lead to a more physically based simulation of aerosol direct and indirect radiative forcings. This study examines the global variation in particle size distribution simulated by twelve global aerosol microphysics models to quantify model diversity and to identify any common biases against observations. Evaluation against size distribution measurements from a new European network of aerosol supersites shows that the mean model agrees quite well with the observations at many sites on the annual mean, but there are some seasonal biases common to many sites. In particular, at many of these European sites, the accumulation mode number concentration is biased low during winter and Aitken mode concentrations tend to be overestimated in winter and underestimated in summer. At high northern latitudes, the models strongly underpredict Aitken and accumulation particle concentrations compared to the measurements, consistent with previous studies that have highlighted the poor performance of global aerosol models in the Arctic. In the marine boundary layer, the models capture the observed meridional variation in the size distribution, which is dominated by the Aitken mode at high latitudes, with an increasing concentration of accumulation particles with decreasing latitude. Considering vertical profiles, the models reproduce the observed peak in total particle concentrations in the upper troposphere due to new particle formation, although modelled peak concentrations tend to be biased high over Europe. Overall, the multi-model-mean dataset simulates the global variation of the particle size distribution with a good degree of skill, suggesting that most of the individual global aerosol microphysics models are performing well, although the large model diversity indicates that some models are in poor agreement with the observations. Further work is required to better constrain size-resolved primary and secondary particle number sources, and an improved understanding of nucleation and growth (e.g. the role of nitrate and secondary organics) will improve the fidelity of simulated particle size distributions.

Previous studies have identified over 23 loci associated with more than 26 urine metabolites levels in adults, which are known as urinary metabolite quantitative trait loci (metabQTLs). The aim of the present study is the identification for the first time of urinary metabQTLs in children and their interaction with dietary patterns. Association between genome-wide genotyping data and 44 urine metabolite levels measured by proton nuclear magnetic resonance spectroscopy was tested in 996 children from the Human Early Life Exposome project. Twelve statistically significant urine metabQTLs were identified, involving 11 unique loci and 10 different metabolites. Comparison with previous findings in adults revealed that six metabQTLs were already known, and one had been described in serum and three were involved the same locus as other reported metabQTLs but had different urinary metabolites. The remaining two metabQTLs represent novel urine metabolite-locus associations, which are reported for the first time in this study [single nucleotide polymorphism (SNP) rs12575496 for taurine, and the missense SNP rs2274870 for 3-hydroxyisobutyrate]. Moreover, it was found that urinary taurine levels were affected by the combined action of genetic variation and dietary patterns of meat intake as well as by the interaction of this SNP with beverage intake dietary patterns. Overall, we identified 12 urinary metabQTLs in children, including two novel associations. While a substantial part of the identified loci affected urinary metabolite levels both in children and in adults, the metabQTL for taurine seemed to be specific to children and interacted with dietary patterns. A large proportion of European biodiversity today depends on habitat provided by low‐intensity farming practices, yet this resource is declining as European agriculture intensifies. Within the European Union, particularly the central and eastern new member states have retained relatively large areas of species‐rich farmland, but despite increased investment in nature conservation here in recent years, farmland biodiversity trends appear to be worsening. Although the high biodiversity value of Central and Eastern European farmland has long been reported, the amount of research in the international literature focused on farmland biodiversity in this region remains comparatively tiny, and measures within the EU Common Agricultural Policy are relatively poorly adapted to support it. In this opinion study, we argue that, 10 years after the accession of the first eastern EU new member states, the continued under‐representation of the low‐intensity farmland in Central and Eastern Europe in the international literature and EU policy is impeding the development of sound, evidence‐based conservation interventions. The biodiversity benefits for Europe of existing low‐intensity farmland, particularly in the central and eastern states, should be harnessed before they are lost. Instead of waiting for species‐rich farmland to further decline, targeted research and monitoring to create locally appropriate conservation strategies for these habitats is needed now. Their continued existence depends on some form of human activity, for either production or conservation purposes, or both. We examined the share of semi-natural grasslands within the general grassland areas in boreal Europe. We reviewed research literature across the region to compile evidence on semi-natural grasslands and other semi-natural habitats, such as wooded pastures, in respect to a range of topics such as ecology, land-use change, socioeconomics, and production. We also explored drivers of the research agenda and outlined future research needs. Challenges are faced when defining and quantifying semi-natural habitats even across a restricted region. Agricultural development and other policies clearly impact the research agenda in various countries. There are recent signs of a shift from classical ecological studies toward more multidisciplinary and integrated research. To sufficiently address the threats faced by semi-natural habitats, political and research frameworks in the European Union should pay more attention to the social-ecological complexity inherent in their management and should support the engagement of various actors into participatory governance processes. This is in line with a full-farm approach implicit in high nature value farming systems. Peer reviewed

Backgroun Ecosystem services (ES) mapping is becoming mainstream in many sustainability assessments, but its impact on real world decision-making is still limited. Robustness, enduser relevance and transparency have been identified as key attributes needed for effective ES mapping. However, these requirements are not always met due to multiple challenges, referred to here as bottlenecks, that scientists, practitioners, policy makers and users from other public and private sectors encounter along the mapping process. A selection of commonly encountered ES mapping bottlenecks that relate to seven themes: i) map-maker map-user interaction ii) nomenclature and ontologies iii) skills and background iv) data and maps availability v) methods-selection vi) technical difficulties and vii) over-simplification of mapping process/output. The authors synthesise the variety of solutions already applied by map-makers and map-users to mitigate or cope with these bottlenecks and discuss the emerging trade-offs amongst different solutions. Tackling the bottlenecks described here is a crucial first step towards more effective ES mapping, which can in turn ensure the adequate impact of ES mapping in decision-making. In MCDM, a crowd as a whole, or its individual members, generally make decisions free from any influence of valence, arousal, emotional state or environment. In contrast, various theories dealing with crowd psychology (Gustave Le Bon, Freudian, Deindividuation, Convergence, Emergent norm, Social identity) analyze, in one form or another, the emotions of the crowd. According to above theories, crowd is influenced by a range of behavioral factors, such as physical, social, psychological, culture, norms, and emotions. It can be argued that the emotional state, valence and arousal of crowds affect their decision making to a considerable degree and multiple criteria crowd behavior modeling must, therefore, consider this impact as well. In this light, the integration of crowd simulation and biometric methods, behavioral operations research and emotions in decision making has taken a prominent place as it leads to a better understanding of crowd emotions and crowd decision making. In this context, the authors developed the Affective Analytics of Demonstration Sites (ANDES) that added to this body of research in four ways. The crowd analysis and simulations conducted with ANDES used a neuro decision matrix. The matrix contains a detailed description of demonstration sites (public spaces) in question and the emotions, valence, arousal and physiological parameters of people present there. With ANDES’s Remote Sensor Network, emotional (emotions, valence, arousal) and physiological (average crowd facial temperature, crowd composition by gender and age group, etc.) parameters of people present at demonstration sites can be mapped. ANDES can assist experts in more effective implementations of public spaces planning and a participation process by attendees by collecting and examining various layers of data on the emotional and physiological parameters of visitors based on a visitors-centric public spaces planning approach. ANDES can determine the public space and real estate values.

Este ctenostomado ha sido considerado por mucho tiempo como una especie nativa del Mar Mediterráneo, pero se ha sugerido recientemente que es originario del Caribe. Es probable que haya sido introducido incrustado en los cascos de los barcos. The spaghetti bryozoan Amathia verticillata, formerly known as Zoobotryon verticillatum, was first described in 1822 from Naples, Italy, although this species was already present in 1807 at Cadiz, Spain. This ctenostome has long been considered a native species in the Mediterranean Sea but it has recently been suggested to be of Caribbean origin. It is most likely to have been introduced by vessels as hull fouling. This pseudo-indigenous species, i.e. a non-indigenous species (NIS) having been perceived to be native, has been found in several marinas and harbours within the Mediterranean Sea. In November 2014, this bryozoan species was abundant in the La Grande Motte marina on the south coast of France. Several thousand colonies were estimated to be present within this marina attached to the floating pontoon units that supported a floating boardwalk. Of the berthed craft examined, 31% were fouled with this species, and it was occasionally a prominent fouling species. Several macroinvertebrate species were associated with A. verticillata colonies, including some NIS, Paracerceis sculpta, Paranthura japonica and Caprella scaura, that are recorded for the first time from the Mediterranean coast of France. A. verticillata might support their transfer elsewhere by providing a habitat and substrate when attached to vessel hulls.

Due to practical reasons, the most suitable positions for modifications are C5 of pyrimidines and C7 of purines. Unfortunately, by using these two positions only, one cannot expand a repertoire of modified nucleotides to a maximum. Here, we demonstrate the synthesis and enzymatic incorporation of novel N-4-acylated 2'-deoxycytidine nucleotides (dC(Acyl)). We find that a variety of family A and B DNA polymerases efficiently use dC(Acyl)TPs as substrates. In addition to the formation of complementary C-Acyl center dot G pair, a strong basepairing between N-4-acyl-cytosine and adenine takes place when Taq, Klenow fragment (exo-), Bsm and KOD XL DNA polymerases are used for the primer extension reactions. In contrast, a proofreading phi29 DNA polymerase successfully utilizes dC(Acyl)TPs but is prone to form C-Acyl center dot A base pair under the same conditions. Moreover, we show that terminal deoxynucleotidyl transferase is able to incorporate as many as several hundred N-4-acylated-deoxycytidine nucleotides. These data reveal novel N-4-acylated deoxycytidine nucleotides as beneficial substrates for the enzymatic synthesis of modified DNA, which can be further applied for specific labelling of DNA fragments, selection of aptamers or photoimmobilization.

This work compares meteorological results from different regional climate model (RCM) implementations in the Mediterranean area, with a focus on the northern Adriatic Sea. The need to use these datasets as atmospheric forcings (wind and atmospheric pressure fields) for coastal hydrodynamic models to assess future changes in the coastal hydrodynamics, is the basis of the presented analysis. It would allow the assessment of uncertainties due to atmospheric forcings in providing coastal current, surge and wave climate changes from future implementations of hydrodynamic models. Two regional climate models, with different spatial resolutions, downscaled from two different global climate models (whose atmospheric components are, respectively, ECHAM4 and ECHAM5), were considered. In particular, the RCM delivered wind and atmospheric pressure fields were compared with measurements at four stations along the Italian Adriatic coast. The analyses were conducted using a past control period, 1960–1990, and the A1B IPCC future scenario (2070–2100). The chosen scenario corresponds to a world of very rapid economic and demographic growth that peaks in mid-century, with a rapid introduction of new efficient technologies, which balance fossil and non-fossil resources (IPCC, 2007). Consideration is given to the accuracy of each model at reproducing the basic statistics and the trends. The role of models' spatial resolution in reproducing global and local scale meteorological processes is also discussed. The Adriatic Sea climate is affected by the orography that produces a strengthening of north-eastern katabatic winds like bora. Therefore, spatial model resolution, both for orography and for a better resolution of coastline (Cavaleri et al., 2010), is one of the important factors in providing more realistic wind forcings for future hydrodynamic models implementations. However, also the characteristics in RCM setup and parameterization can explain differences between the datasets. The analysis from an ensemble of model implementation would provide more robust indications on climatic wind and atmospheric pressure variations. The scenario-control comparison shows a general increase in the mean atmospheric pressure values while a decrease in mean wind speed and in extreme wind events is seen, particularly for the datasets with higher spatial resolution.

Here, we aimed to describe the early-life exposome. Specifically, we focused on the correlations between multiple environmental exposures, their patterns and their variability across European regions and across time (pregnancy and childhood periods). We relied on the Human Early-Life Exposome (HELIX) project, in which 87 environmental exposures during pregnancy and 122 during the childhood period (grouped in 19 exposure groups) were assessed in 1301 pregnant mothers and their children at 6-11 years in 6 European birth cohorts. Some correlations between exposures in the same exposure group reached high values above 0.8. The median correlation within exposure groups was >0.3 for many exposure groups, reaching 0.69 for water disinfection by products in pregnancy and 0.67 for the meteorological group in childhood. Median correlations between different exposure groups rarely reached 0.3. Some correlations were driven by cohort-level associations (e.g. air pollution and chemicals). Ten principal components explained 45% and 39% of the total variance in the pregnancy and childhood exposome, respectively, while 65 and 90 components were required to explain 95% of the exposome variability. Correlations between maternal (pregnancy) and childhood exposures were high (>0.6) for most exposures modeled at the residential address (e.g. air pollution), but were much lower and even close to zero for some chemical exposures. In conclusion, the early life exposome was high dimensional, meaning that it cannot easily be measured by or reduced to fewer components. Correlations between exposures from different exposure groups were much lower than within exposure groups, which have important implications for co-exposure confounding in multiple exposure studies. Also, we observed the early life exposome to be variable over time and to vary by cohort, so measurements at one time point or one place will not capture its complexities. Many of the next generation of global climate models will include aerosol schemes which explicitly simulate the microphysical processes that determine the particle size distribution. These models enable aerosol optical properties and cloud condensation nuclei (CCN) concentrations to be determined by fundamental aerosol processes, which should lead to a more physically based simulation of aerosol direct and indirect radiative forcings. This study examines the global variation in particle size distribution simulated by twelve global aerosol microphysics models to quantify model diversity and to identify any common biases against observations. Evaluation against size distribution measurements from a new European network of aerosol supersites shows that the mean model agrees quite well with the observations at many sites on the annual mean, but there are some seasonal biases common to many sites. In particular, at many of these European sites, the accumulation mode number concentration is biased low during winter and Aitken mode concentrations tend to be overestimated in winter and underestimated in summer. At high northern latitudes, the models strongly underpredict Aitken and accumulation particle concentrations compared to the measurements, consistent with previous studies that have highlighted the poor performance of global aerosol models in the Arctic. In the marine boundary layer, the models capture the observed meridional variation in the size distribution, which is dominated by the Aitken mode at high latitudes, with an increasing concentration of accumulation particles with decreasing latitude. Considering vertical profiles, the models reproduce the observed peak in total particle concentrations in the upper troposphere due to new particle formation, although modelled peak concentrations tend to be biased high over Europe. Overall, the multi-model-mean dataset simulates the global variation of the particle size distribution with a good degree of skill, suggesting that most of the individual global aerosol microphysics models are performing well, although the large model diversity indicates that some models are in poor agreement with the observations. Further work is required to better constrain size-resolved primary and secondary particle number sources, and an improved understanding of nucleation and growth (e.g. the role of nitrate and secondary organics) will improve the fidelity of simulated particle size distributions.

Previous studies have identified over 23 loci associated with more than 26 urine metabolites levels in adults, which are known as urinary metabolite quantitative trait loci (metabQTLs). The aim of the present study is the identification for the first time of urinary metabQTLs in children and their interaction with dietary patterns. Association between genome-wide genotyping data and 44 urine metabolite levels measured by proton nuclear magnetic resonance spectroscopy was tested in 996 children from the Human Early Life Exposome project. Twelve statistically significant urine metabQTLs were identified, involving 11 unique loci and 10 different metabolites. Comparison with previous findings in adults revealed that six metabQTLs were already known, and one had been described in serum and three were involved the same locus as other reported metabQTLs but had different urinary metabolites. The remaining two metabQTLs represent novel urine metabolite-locus associations, which are reported for the first time in this study [single nucleotide polymorphism (SNP) rs12575496 for taurine, and the missense SNP rs2274870 for 3-hydroxyisobutyrate]. Moreover, it was found that urinary taurine levels were affected by the combined action of genetic variation and dietary patterns of meat intake as well as by the interaction of this SNP with beverage intake dietary patterns. Overall, we identified 12 urinary metabQTLs in children, including two novel associations. While a substantial part of the identified loci affected urinary metabolite levels both in children and in adults, the metabQTL for taurine seemed to be specific to children and interacted with dietary patterns.
