    Macrophages play important roles in conditions ranging from host immune defense to tissue regeneration and polarize their functional phenotype accordingly. Next to differences in the use of L-arginine and the production of different cytokines, inflammatory M1 macrophages and anti-inflammatory M2 macrophages are also metabolically distinct. In mammals, M1 macrophages show metabolic reprogramming toward glycolysis, while M2 macrophages rely on oxidative phosphorylation to generate energy. The presence of polarized functional immune phenotypes conserved from mammals to fish led us to hypothesize that a similar metabolic reprogramming in polarized macrophages exists in carp. We studied mitochondrial function of M1 and M2 carp macrophages under basal and stressed conditions to determine oxidative capacity by real-time measurements of oxygen consumption and glycolytic capacity by measuring lactate-based acidification. In M1 macrophages, we found increased nitric oxide production and irg1 expression in addition to altered oxidative phosphorylation and glycolysis. In M2 macrophages, we found increased arginase activity, and both oxidative phosphorylation and glycolysis were similar to control macrophages. These results indicate that M1 and M2 carp macrophages show distinct metabolic signatures and indicate that metabolic reprogramming may occur in carp M1 macrophages. This immunometabolic reprogramming likely supports the inflammatory phenotype of polarized macrophages in teleost fish such as carp, similar to what has been shown in mammals.

    Several chemical reactions related to the oxidation and combustion of sulfur are investigated using a number of computational chemistry methods with the objective of determining appropriate methods for use in developing an elementary reaction mechanism for oxidation of sulfur. Calculations are focused on thermochemical properties and reaction energetics for reactive species and transition state structures for reactions in the oxidation / combustion of sulfur. Reactions involving several intermediates resulting from the reactions of S2 with oxygen were investigated with the density functional theory B3LYP (with several basis sets) and BB1K/GTLarge. The composite ab-initio methods G2, G3, G3MP2, G3B3, G3MP2B3 and CBS-QB3 were also used. Enthalpies of a series of sulfur compounds and transition state structures are calculated using the ab initio and DFT calculations. The calculations were combined with isodesmic reaction analysis, whenever possible, in order to cancel error and improve the accuracy of the calculations. Results show that all B3LYP DFT calculations including the 6-311++G(3df,2p) basis set show poor outcome in estimating the enthalpy of reactions involving S2. The six composite methods have all shown consistency with each other and their calculated reaction energies / bond energies are in good agreement with the available literature. Kinetic parameters for calculation of the kinetic parameters on SO3 dissociation to SO2 and O using the canonical transition state theory are reported.

    In future power systems, widespread small-scale energy storage systems (ESSs) can be aggregated to provide ancillary services. In this context, a new load frequency control scheme which incorporates the energy storage aggregator (ESA) and its associated disturbance observer is proposed. The disturbance observer is designed to supplement the secondary frequency control for the ESA, therefore the system frequency response and recovery can be improved. Within the ESA, a finite-time leader-follower consensus algorithm is proposed to control the small-scale ESSs via sparse communication networks. This algorithm ensures that the ESA tracks the frequency control signals, while the state-of-charge among each ESS is balanced in finite-time. The external characteristics of the ESA will resemble to that of one large-scale ESS. Numerical examples demonstrate the convergence of the ESA under different communication graphs. The effectiveness of the entire scheme for power system frequency control is validated under a variety of scenarios that include contingency and normal operation.     We propose novel positive numerical integrators for approximating predator-prey models. The schemes are based on suitable symplectic procedures applied to the dynamical system written in terms of the log transformation of the original variables. Even if this approach is not new when dealing with Hamiltonian systems, it is of particular interest in population dynamics since the positivity of the approximation is ensured without any restriction on the temporal step size. When applied to separable M-systems, the resulting schemes are proved to be explicit, positive, Poisson maps. The approach is generalized to predator-prey dynamics which do not exhibit an M-system structure and successively to reaction-diffusion equations describing spatially extended dynamics. A classical polynomial Krylov approximation for the diffusive term joint with the proposed schemes for the reaction, allows us to propose numerical schemes which are explicit when applied to well established ecological models for predator-prey dynamics. Numerical simulations show that the considered approach provides results which outperform the numerical approximations found in recent literature.

    AbstractIn this paper, we introduce and test a framework to qualitatively assess the environmental impact of climate adaptation innovations with the ambition to facilitate the implementation of these adaptations. The framework was designed to enable continuous environmentally conscious benchmarking based on three environmental performance indicators: sustainable design, environmental impact and ecological impact. It was pilot tested by uninvolved experts and key-persons for two large-scale nature-based flood adaptation innovations in the Netherlands and discussed with environmental assessment professionals. Our findings indicate how the inclusion of our framework helps to identify important knowledge gaps regarding environmental co-benefits and trade-offs, and can be beneficial to both those developing the innovation and the local authorities charged with assessing the suitability of innovations. We conclude by noting how the incorporation of environmental impact assessment from the design stage of adaptations could supplement existing environmental assessment regulations pre-empting concerns rather than reacting to them.

    En la presente tesis doctoral se han desarrollado modelos para estimar la distribución vertical de la carga de combustible disponible del dosel arbóreo, tanto a partir de datos LiDAR como de variables de rodal medidas en campo. Además, se han obtenido modelos de estimación de variables del combustible de superficie y del dosel arbóreo a partir de imágenes del satélite Sentinel-2A, que permiten generar cartografía del riesgo potencial de fuego de copas. Por otro lado, se ha evaluado el efecto a medio plazo del tratamiento de clara por si sola, sin intervención en los combustibles de superficie, sobre el complejo de combustible y el comportamiento y la severidad potenciales del fuego, así como la capacidad de los rodales quemados para proteger el suelo contra la erosión post-incendio. Todos estos estudios se realizaron en pinares del noroeste de España y los resultados obtenidos aportan nueva información sobre combustibles forestales, conectándola con el comportamiento esperado del fuego en incendios, siendo de gran utilidad no sólo en la gestión del fuego forestal sino también en la gestión forestal en general.

    A fox circovirus was identified in serum samples from foxes with unexplained neurologic signs by using viral metagenomics. Fox circovirus nucleic acid was localized in histological lesions of the cerebrum by in situ hybridization. Viruses from the family Circoviridae may have neurologic tropism more commonly than previously anticipated.

    International audience; Die Reduktion von Pflanzenfraß durch Insekten ist eine der Leistungen, die von der Baumdiversität in Waldökosystemen erbracht wird. Während zunehmend anerkannt wird, dass die Zusammensetzung von Baumgemeinschaften eine wichtige Rolle beim Auslösen von gemeinschaftlicher Abwehr gegen Herbivore spielt, ist über die zugrunde liegenden Mechanismen wenig bekannt. Wir untersuchten diese Frage im ORPHEE-Experiment, indem wir den Befall durch den Pinienprozessionsspinner (Thaumetopoea pityocampa) über einen Gradienten von Monokultur bis zur Fünf-Arten-Mischung registrierten. Wir zeigten, dass der Baum-Artenreichtum per se keinen Einfluss auf die Befallswahrscheinlichkeit hatte. Dagegen hingen die Befallsraten stark von der Zusammensetzung der Bestände ab. Systeme von Pinie (Pinus pinaster) und Birke (Betula pendula) wurden weniger stark befallen, während Mischungen von Pinien und Eichen (Quercus spp) häufiger als Reinbestände von Pinien attackiert wurden. Wenn man die relative Höhe von Pinien und der vergesellschafteten Laubbaumarten in Betracht zieht, könnte dieser Effekt mit der Apparenz der Pinien erklärt werden. Wir meinen deshalb, dass die verminderte Apparenz der Pinien visuelle Signale stört, die von den Weibchen genutzt werden, um vor der Eiablage Wirtsbäume auszuwählen. Diese Untersuchung macht deutlich, dass es notwendig ist, Eigenschaften wie die Wachstumsgeschwindigkeit zu berücksichtigen, wenn die Baumarten für eine Mischkultur ausgewählt werden, um die Abwehr eines Forstbestandes gegen Schadinsekten zu verbessern.; The reduction of insect herbivory is one of the services provided by tree diversity in forest ecosystems. While it is increasingly acknowledged that the compositional characteristics of tree species assemblages play a major role in triggering associational resistance to herbivores, underlying mechanisms are less well known. We addressed this question in the ORPHEE experiment by assessing pine processionary moth infestations (Thaumetopoea pityocampa) across a tree diversity gradient from pine monocultures to five species mixtures. We showed that tree species richness per se had no effect on the probability of attack by this pest. By contrast, the infestation rate was strongly dependent on plot composition. Mixtures of pines (Pinus pinaster) and birches (Betula pendula) were less prone to T. pityocampa infestations, whereas mixtures of pines and oaks (Quercus spp.) were more often attacked than pine monocultures. By taking into account the relative height of pines and associated broadleaved species, this effect could be explained by pine apparency. Pines were on average 343 ± 5 cm height. Birches, as fast growing trees, were slightly taller than pines (363 ± 6 cm), while oak trees were significantly smaller (74 ± 1 cm). Host trees of T. pityocampa were then partly hidden in mixtures of pines and birches but more apparent in mixtures with oaks. We suggest that reduced pine apparency disrupted visual cues used by female moths to select host trees prior to oviposition. This study highlights the need to take into account tree traits such as growth rate when selecting the tree species that have to be associated in order to improve forest resistance to pest insects.

    Personalised nutrition (PN) has the potential to reduce disease risk and optimise health and performance. Although previous research has shown good acceptance of the concept of PN in the UK, preferences regarding the delivery of a PN service (e.g. online v. face-to-face) are not fully understood. It is anticipated that the presence of a free at point of delivery healthcare system, the National Health Service (NHS), in the UK may have an impact on end-user preferences for deliverances. To determine this, supplementary analysis of qualitative data obtained from focus group discussions on PN service delivery, collected as part of the Food4Me project in the UK and Ireland, was undertaken. Irish data provided comparative analysis of a healthcare system that is not provided free of charge at the point of delivery to the entire population. Analyses were conducted using the ‘framework approach’ described by Rabiee (Focus-group interview and data analysis. Proc Nutr Soc 63, 655-660). There was a preference for services to be led by the government and delivered face-to-face, which was perceived to increase trust and transparency, and add value. Both countries associated paying for nutritional advice with increased commitment and motivation to follow guidelines. Contrary to Ireland, however, and despite the perceived benefit of paying, UK discussants still expected PN services to be delivered free of charge by the NHS. Consideration of this unique challenge of free healthcare that is embedded in the NHS culture will be crucial when introducing PN to the UK.

    Macrophages play important roles in conditions ranging from host immune defense to tissue regeneration and polarize their functional phenotype accordingly. Next to differences in the use of L-arginine and the production of different cytokines, inflammatory M1 macrophages and anti-inflammatory M2 macrophages are also metabolically distinct. In mammals, M1 macrophages show metabolic reprogramming toward glycolysis, while M2 macrophages rely on oxidative phosphorylation to generate energy. The presence of polarized functional immune phenotypes conserved from mammals to fish led us to hypothesize that a similar metabolic reprogramming in polarized macrophages exists in carp. We studied mitochondrial function of M1 and M2 carp macrophages under basal and stressed conditions to determine oxidative capacity by real-time measurements of oxygen consumption and glycolytic capacity by measuring lactate-based acidification. In M1 macrophages, we found increased nitric oxide production and irg1 expression in addition to altered oxidative phosphorylation and glycolysis. In M2 macrophages, we found increased arginase activity, and both oxidative phosphorylation and glycolysis were similar to control macrophages. These results indicate that M1 and M2 carp macrophages show distinct metabolic signatures and indicate that metabolic reprogramming may occur in carp M1 macrophages. This immunometabolic reprogramming likely supports the inflammatory phenotype of polarized macrophages in teleost fish such as carp, similar to what has been shown in mammals.

    Several chemical reactions related to the oxidation and combustion of sulfur are investigated using a number of computational chemistry methods with the objective of determining appropriate methods for use in developing an elementary reaction mechanism for oxidation of sulfur. Calculations are focused on thermochemical properties and reaction energetics for reactive species and transition state structures for reactions in the oxidation / combustion of sulfur. Reactions involving several intermediates resulting from the reactions of S2 with oxygen were investigated with the density functional theory B3LYP (with several basis sets) and BB1K/GTLarge. The composite ab-initio methods G2, G3, G3MP2, G3B3, G3MP2B3 and CBS-QB3 were also used. Enthalpies of a series of sulfur compounds and transition state structures are calculated using the ab initio and DFT calculations. The calculations were combined with isodesmic reaction analysis, whenever possible, in order to cancel error and improve the accuracy of the calculations. Results show that all B3LYP DFT calculations including the 6-311++G(3df,2p) basis set show poor outcome in estimating the enthalpy of reactions involving S2. The six composite methods have all shown consistency with each other and their calculated reaction energies / bond energies are in good agreement with the available literature. Kinetic parameters for calculation of the kinetic parameters on SO3 dissociation to SO2 and O using the canonical transition state theory are reported.

    In future power systems, widespread small-scale energy storage systems (ESSs) can be aggregated to provide ancillary services. In this context, a new load frequency control scheme which incorporates the energy storage aggregator (ESA) and its associated disturbance observer is proposed. The disturbance observer is designed to supplement the secondary frequency control for the ESA, therefore the system frequency response and recovery can be improved. Within the ESA, a finite-time leader-follower consensus algorithm is proposed to control the small-scale ESSs via sparse communication networks. This algorithm ensures that the ESA tracks the frequency control signals, while the state-of-charge among each ESS is balanced in finite-time. The external characteristics of the ESA will resemble to that of one large-scale ESS. Numerical examples demonstrate the convergence of the ESA under different communication graphs. The effectiveness of the entire scheme for power system frequency control is validated under a variety of scenarios that include contingency and normal operation.     We propose novel positive numerical integrators for approximating predator-prey models. The schemes are based on suitable symplectic procedures applied to the dynamical system written in terms of the log transformation of the original variables. Even if this approach is not new when dealing with Hamiltonian systems, it is of particular interest in population dynamics since the positivity of the approximation is ensured without any restriction on the temporal step size. When applied to separable M-systems, the resulting schemes are proved to be explicit, positive, Poisson maps. The approach is generalized to predator-prey dynamics which do not exhibit an M-system structure and successively to reaction-diffusion equations describing spatially extended dynamics. A classical polynomial Krylov approximation for the diffusive term joint with the proposed schemes for the reaction, allows us to propose numerical schemes which are explicit when applied to well established ecological models for predator-prey dynamics. Numerical simulations show that the considered approach provides results which outperform the numerical approximations found in recent literature.

    AbstractIn this paper, we introduce and test a framework to qualitatively assess the environmental impact of climate adaptation innovations with the ambition to facilitate the implementation of these adaptations. The framework was designed to enable continuous environmentally conscious benchmarking based on three environmental performance indicators: sustainable design, environmental impact and ecological impact. It was pilot tested by uninvolved experts and key-persons for two large-scale nature-based flood adaptation innovations in the Netherlands and discussed with environmental assessment professionals. Our findings indicate how the inclusion of our framework helps to identify important knowledge gaps regarding environmental co-benefits and trade-offs, and can be beneficial to both those developing the innovation and the local authorities charged with assessing the suitability of innovations. We conclude by noting how the incorporation of environmental impact assessment from the design stage of adaptations could supplement existing environmental assessment regulations pre-empting concerns rather than reacting to them.

    En la presente tesis doctoral se han desarrollado modelos para estimar la distribución vertical de la carga de combustible disponible del dosel arbóreo, tanto a partir de datos LiDAR como de variables de rodal medidas en campo. Además, se han obtenido modelos de estimación de variables del combustible de superficie y del dosel arbóreo a partir de imágenes del satélite Sentinel-2A, que permiten generar cartografía del riesgo potencial de fuego de copas. Por otro lado, se ha evaluado el efecto a medio plazo del tratamiento de clara por si sola, sin intervención en los combustibles de superficie, sobre el complejo de combustible y el comportamiento y la severidad potenciales del fuego, así como la capacidad de los rodales quemados para proteger el suelo contra la erosión post-incendio. Todos estos estudios se realizaron en pinares del noroeste de España y los resultados obtenidos aportan nueva información sobre combustibles forestales, conectándola con el comportamiento esperado del fuego en incendios, siendo de gran utilidad no sólo en la gestión del fuego forestal sino también en la gestión forestal en general.

    A fox circovirus was identified in serum samples from foxes with unexplained neurologic signs by using viral metagenomics. Fox circovirus nucleic acid was localized in histological lesions of the cerebrum by in situ hybridization. Viruses from the family Circoviridae may have neurologic tropism more commonly than previously anticipated.

    International audience; Die Reduktion von Pflanzenfraß durch Insekten ist eine der Leistungen, die von der Baumdiversität in Waldökosystemen erbracht wird. Während zunehmend anerkannt wird, dass die Zusammensetzung von Baumgemeinschaften eine wichtige Rolle beim Auslösen von gemeinschaftlicher Abwehr gegen Herbivore spielt, ist über die zugrunde liegenden Mechanismen wenig bekannt. Wir untersuchten diese Frage im ORPHEE-Experiment, indem wir den Befall durch den Pinienprozessionsspinner (Thaumetopoea pityocampa) über einen Gradienten von Monokultur bis zur Fünf-Arten-Mischung registrierten. Wir zeigten, dass der Baum-Artenreichtum per se keinen Einfluss auf die Befallswahrscheinlichkeit hatte. Dagegen hingen die Befallsraten stark von der Zusammensetzung der Bestände ab. Systeme von Pinie (Pinus pinaster) und Birke (Betula pendula) wurden weniger stark befallen, während Mischungen von Pinien und Eichen (Quercus spp) häufiger als Reinbestände von Pinien attackiert wurden. Wenn man die relative Höhe von Pinien und der vergesellschafteten Laubbaumarten in Betracht zieht, könnte dieser Effekt mit der Apparenz der Pinien erklärt werden. Die Pinien waren im Mittel 343 ± 5 cm hoch. Die Birken waren als schnellwachsende Bäume etwas höher 363 ± 6 cm) als die Pinien, während die Eichen bedeutend kleiner waren (74 ± 1 cm). Die Wirtsbäume von T. pityocampa waren damit in den Mischbeständen von Pinie und Birke teilweise verborgen, traten bei Vergesellschaftung mit Eichen aber deutlicher in Erscheinung. Wir meinen deshalb, dass die verminderte Apparenz der Pinien visuelle Signale stört, die von den Weibchen genutzt werden, um vor der Eiablage Wirtsbäume auszuwählen. Diese Untersuchung macht deutlich, dass es notwendig ist, Eigenschaften wie die Wachstumsgeschwindigkeit zu berücksichtigen, wenn die Baumarten für eine Mischkultur ausgewählt werden, um die Abwehr eines Forstbestandes gegen Schadinsekten zu verbessern.; The reduction of insect herbivory is one of the services provided by tree diversity in forest ecosystems. While it is increasingly acknowledged that the compositional characteristics of tree species assemblages play a major role in triggering associational resistance to herbivores, underlying mechanisms are less well known. We addressed this question in the ORPHEE experiment by assessing pine processionary moth infestations (Thaumetopoea pityocampa) across a tree diversity gradient from pine monocultures to five species mixtures. We showed that tree species richness per se had no effect on the probability of attack by this pest. By contrast, the infestation rate was strongly dependent on plot composition. Mixtures of pines (Pinus pinaster) and birches (Betula pendula) were less prone to T. pityocampa infestations, whereas mixtures of pines and oaks (Quercus spp.) were more often attacked than pine monocultures. By taking into account the relative height of pines and associated broadleaved species, this effect could be explained by pine apparency. Pines were on average 343 ± 5 cm height. Birches, as fast growing trees, were slightly taller than pines (363 ± 6 cm), while oak trees were significantly smaller (74 ± 1 cm). Host trees of T. pityocampa were then partly hidden in mixtures of pines and birches but more apparent in mixtures with oaks. We suggest that reduced pine apparency disrupted visual cues used by female moths to select host trees prior to oviposition. This study highlights the need to take into account tree traits such as growth rate when selecting the tree species that have to be associated in order to improve forest resistance to pest insects.

    Personalised nutrition (PN) has the potential to reduce disease risk and optimise health and performance. Although previous research has shown good acceptance of the concept of PN in the UK, preferences regarding the delivery of a PN service (e.g. online v. face-to-face) are not fully understood. It is anticipated that the presence of a free at point of delivery healthcare system, the National Health Service (NHS), in the UK may have an impact on end-user preferences for deliverances. To determine this, supplementary analysis of qualitative data obtained from focus group discussions on PN service delivery, collected as part of the Food4Me project in the UK and Ireland, was undertaken. Irish data provided comparative analysis of a healthcare system that is not provided free of charge at the point of delivery to the entire population. Analyses were conducted using the ‘framework approach’ described by Rabiee (Focus-group interview and data analysis. Proc Nutr Soc 63, 655-660). There was a preference for services to be led by the government and delivered face-to-face, which was perceived to increase trust and transparency, and add value. Both countries associated paying for nutritional advice with increased commitment and motivation to follow guidelines. Contrary to Ireland, however, and despite the perceived benefit of paying, UK discussants still expected PN services to be delivered free of charge by the NHS. Consideration of this unique challenge of free healthcare that is embedded in the NHS culture will be crucial when introducing PN to the UK.
